Study on in Vitro Propagation of Tea (camellia Sinensis L.)

Abstract/ Executive Summary:

Shoot tip explants of tea (Camellia sinensis L.) clone BT1, BT2, BT4, BT5, BT6, BT7, BT8, BT10 and BT11 were cultured in vitro. Regeneration of plantlets was observed in MS media supplemented with 1 mg/lKn + 1 mg/IIAA, 1.5 mg/I Kn + 1.5 mg/I IAA and 1 mg/I BAP + 1 mg/I IAA. Plantlets obtained in regenerating media were subsequently transferred to rooting media containing several combinations of 1/2 MS, IBA and IAA but no root initiation was observed. Stem segments of clone BTl, BT2, BT5, BT6 and BT10 were cultured aseptically in vitro. Callus was obtained when MS media were supplemented with 2mg/I 2,4-0, 1 mg/I 2,4-0 + 300CH, 1.5mg/l 2,4-0 + 300CH. Higher concentrations of 2,4-0 were found to inhibit callus induction. The calli thus obtained were transferred to regenerating media containing several combinations of BAP, NAA and Kinetin but no organo genesis was observed.

Objectives:

To develop suitable protocol for in vitro propagation of elite clones.

Methodology:

Shoot tips and stem segments of clone BT1, BT2, BT4, BT5, BT6, BT7, BT8, BT10 and BT11 were taken as source of in vitro propagation. Shoot tips about 5mm were cut. The explants were surface sterilized by successive treatments with 70% ethanol for 2 minutes followed by the treatment of 0.2% mercuric chloride solution for 15 minutes. Then the treated explants were rinsed with sterile distilled water 4-5 times and aseptically placed in 30 x 150 mm test tubes each containing 15ml of the MS medium with different concentration of hormone supplements. On the other hand, stem segments were collected from the growing shoots of tea plants (between the 4^th - 8^th leaf) and were cut into small pieces, the length of which was about 2mm. The explants were then surface sterilized by agitating with a solution of 7% calcium hypochloride and 0.01 % Tween-20 for 15 minutes. Then the explants were rinsed in sterile distilled water changing it 4-5 times. The explants were placed in MS medium with different combination of growth hormones.

Achievement/Findings:

Stem segments of clone BTl, BT2, BT5, BT6 and BT10 were found to induce calli but the percentage of callus induction was not identical in all the clones studied. High concentration of 2,4-0 was found to inhibit callus induction. Callus induction from stem segments of tea was reported by several workers.
