Nitrogen Fixation and Respiratory Activity of Azospirillum Spp. Isolated from Saline Habitat of Bangladesh

Abstract/ Executive Summary:

The respiratory activity and nitrogen fixing potential of five strains of Azospirillum lipoferum, three strains of Azospirillum brasilense, three strains of Azospirillum lialopraeferens and one strain of Azospirillum antazonense isolated from saline habitat in the district Satkhira, Bangladesh were studied. The salt, NaCI has shown definite effect on the respiratory activity and nitrogen fixing potential of the selected strains of Azospirillum spp. The activity and the rate of nitrogen fixation of the strains MR-l, MR-3, MR-4, MR-8 and MR-ll were found maximum at 3% NaCI. The other strains MR-5, MR-6, MR-7, MR-13, MR-14, MR-15 and MR-16 showed highest nitrogen fixation and respiratory activity at 5% NaCI. Among the strains, MR-7, MR-13 and MR-16 strains were comparatively more efficient in nitrogen fixing ability and respiratory activity under stressed condition due to salinity.

Objectives:

To study the interaction between salinity stressed condition and nitrogen fixing ability of Azospirillum for developing Azospirillum inoculant for agriculture in saline soil.

Methodology:

Twelve isolates of Azospirillum from saline habitat in the district Satkhira of Bangladesh were thoroughly characterized and identified according to Bergey's Mammal of Determinative Bacteriology. Respiratory activity of the selected strains at different concentrations (0.0, 1.0, 2.0, 3.0, 4.0, 5.0, 6.0 7.0 and 8.0%) of NaCI was determined by measuring the amount of CO₂ evolved by the culture due to respiration. Carbon dioxide evolution was estimated. Nitrogen fixation was determined in terms of the quantity of nitrogen gained in the 72-h-old culture of each strain grown in 25 ml semi-solid nitrogen free malate medium (without bromothymol blue) containing different concentrations (0.0, 1.0, 2.0, 3.0, 4.0, 5.0, 6.0 and 7.0%) of NaCI. The total nitrogen in culture was estimated.

Achievement/Findings:

From statistical analysis it was found that different concentrations of NaCI influenced significantly the nitrogen fixing ability and respiratory activity of the selected strains of Azospirillum spp. Significant positive correlation (r = 0.54) between nitrogen fixation and respiratory activity has been found and this seems to be logical as respiration provides energy for nitrogen fixation. This study indicates that halophilic Azospirillum from saline habitat might be very useful to obtain inoculant of efficient strains for the use in nutrient management programme to increase crop yield in saline agro-ecosystem. The activity and the rate of nitrogen fixation of the strains MR-l, MR-3, MR-4, MR-8 and MR-ll were found maximum at 3% NaCI.
