Response of Mungbean (vigna Radiata (l.) Wilczek) to Inoculation with Bradyrhizobium as Affected by Phosphorus Levels

Abstract/ Executive Summary:

A field experiment was conducted at the farm of Rajbari Testing Station, BARI, Dinajpur during Kharif season of 1989 to study the response of inoculation with Bradyrhizobium inoculants incorporating BINA 403, BINA 407, RCR 3824 and RCR 3825 strains as single and mixed culture and 4 levels of phosphorus ( @ 0, 30, 60 and 90 kg P₂O₅/ha from triple super phosphate) with a basal dose of potassium (@ 30 kg, K₂O/ha from muriate of potash) on growth, root nodulation, yield and yield contributing characters and protein and phosphorus content of mungbean. Bradyrhizobium inoculation increased significantly the number of nodules, nodule weight, shoot length and weight, grain, hay and total protein yield, grain and hay phosphorus content of mungbean compared to uninoculated controls. Differences were found among the cultures, Local mixed culture being the best at nearly all the levels of phospsatic fertilizer in respect of nodulation, grain, hay and totaI protein yield and other character studied. Local single culture was superior to Local exotic mixed culture and Exotic mixed culture was superior to Exotic single culture in almost all the parameters studied. Phosphorus application upto 60 kg P₂O₅/ha progressively and significantly increased nodulation, shoot length and weight, grain, hay and total protein yield and grain and hay phosphorus content. Higher dose of phosphorus decreased the grain and hay Yield and other parameters studied. Bradyrhizobium and phosphorus interaction increased nodulation, nodule weight, shoot weight, grain and hay yield significantly. Highest nodule weight, shoot length and weight, grain, hay and total protein yield and grain and hay phophorus content were obtained with local mixed culture at 60 kg P₂O₅/ha The cultures were also found to increase the fertilizer efficiency in increasing crop yield. The present study also showed that Bradyrhizobium inoculation gave better yield performances at different levels of phosphatic fertilizer and did best at 60 kg P₂O₅/ha.

Objectives:

To conducted a field experiment with single and multistrain rhizobial inoculants under different phosphate levels on the effectiveness and competitiveness of the inoculant as regard to mungbean production.

Methodology:

Two locally isolated (BINA 403 and BINA 407) and two exotic Bradumizoaium strains (RCR 3824 and RCR 3825) were grown separately in yeast extract mannitol broth in 500 ml conical flasks for 5 days on a rotary shaker. Mixed cultures were prepared by mixing equal quantity of fully grown individual culture broth (viable cells in the order of 109/ml broth) in a sterile conical flask. The single strains used were BINA 403 designated as local single culture and RCR 3824 designated as Exotic single culture. The multistrain inoculants were (1) Local mixed culture consisted of BINA 403 and BINA 407, (2) Exotic mixed culture consisted of RCR 3824 and RCR 3825 and (3) Local exotic mixed culture consisted of BINA 403 and RCR 3824 applied as a 1:1 aqueous mixture. Thus there were 6 inoculation treatments viz. 2 single culture, 3 mixed culture and 1 uninoculated control. The field experiment was laid out in triplicated split plot factorial design with phosphorus levels in main plots and inoculation treatments in subplots. The size of each unit plot was 10 sq. m. The crop was grown during kharif season of 1989 (March-May). Phosphorus fertilizer as per plan and a basal dose of muriate of potash @30 kg K₂O/ha were applied before one day of sowing. The inoculated seeds were sown on the furrow which were covered by soils immediately. The line to line distance was maintained 25 cm with plant to plant distance of 10 cm in the line. At 45 days of plant growth 5 randomly selected plants from each plot were carefully uprooted so that no nodule was left in the soil. The whole root system was washed and nodules were separated. Both the shoot and nodules were first air dried, then oven dried at 65° C for 72 hours and their oven dry weights were recorded. The crop was finally harvested at physiological maturity. Nitrogen content of grain and straw samples were estimated by the standard microkjeldahl method. For P content plants samples (grain and straw separately) were digested with triacid mixture and then P was estimated by vanado-molybdophos phoric acid yellow color method using Barton's reagent.

Achievement/Findings:

Results indicated that the inoculation with Rhizobium would be beneficial in mungbean cultivation. Local mixed cultures did better significantly in most of the growth and yield parameters studied. Local single culture was superior to Local exotic mixed culture and Exotic mixed culture was superior to Exotic single culture Bradyrhizobium and phosphorus interaction increased nodulation, nodule weight, shoot weight, grain and hay yield significantly. Highest nodule weight, shoot length and weight, grain, hay and total protein yield and grain and hay phosphorus content were obtained with Local mixed culture at 60 kg P₂O₅/ha. The cultures were also found to increase the fertilizer efficiency in increasing crop yield. The present study also showed that Bradyrhizobium inoculation gave better yield performances at different levels of phosphatic fertilizer and did best at 60 kg P₂O₅/ha.The mixed culture improved the nodule formation in soybean and local strains proved as effective as imported cultures. The exotic strains lost their nodulation ability over a period of three years compared to local strains of Rhizobium in soybean. The mixed rhizobium cultures were significantly superior to single culture in lentil, chickpea and groundnut.
