Cryopreservation of Sperm of Indian Major Carps (catla Catla, Labeo Rohita, Cirrhinus Cirrhosus) for Their Genetically Improved Seed Production.

Abstract/ Executive Summary:

For development and standardization of cryopreservation protocols for Indian major carps, C. catla, L. rohita and C. cirrhosus sperm, six extenders such as egg-yolk citrate, urea egg-yolk, Alsevers solution, Kurokura-2, Ma and Mb and five cryoprotectants viz. DMSO, DMA, glycerol, methanol and ethanol were used. In L. rohita, egg-yolk citrate with DMSO and ethanol, Alsevers solution and urea egg-yolk with DMSO showed ≥80% post-thaw motility. In case of C. catla and C. cirrhosus, Alsevers solution with 10% DMSO showed the best performance and it produced 80±10% and 86±4.2% post-thaw spermatozoan motility respectively. The other two extenders, egg-yolk-citrate and urea egg-yolk along with 10% DMSO produced 76±6.52 and 75±5.0% post-thaw motility for C. catla and 73±4.47% and 75±11.18% post thaw motility for C. cirrhosus respectively. The effect of storage time on the motility of preserved sperm of C. cirrhosus, L. rohita and C. catla were investigated for a period of 90, 130 and 150 days respectively and a gradual reduction in the motility of sperm has been found with the lapse of time. Several cryodiluents were used to preserve the sperm for this experiment and at the end of the storage period 22.2-29.4% motility reduction was recorded for all three IMCs. For sustainable use of cryopreserved sperm, an attempt was taken to determine the suitable egg-sperm ratio during fertilization by using variable number of straws (e.g. 5, 10 and 15 straws) against the approximately equal number of eggs, and 10 straws produced significantly higher number of hatchling compared to others. In breeding trials fertilization of eggs using cryopreserved sperm has been possible for all three species but the percentages of hatching was not consistent at all the time which demands more investigation on development of techniques for thawing of preserved sperm, fertilization of eggs and incubation. When compared the growth of fry produced from cryopreserved and fresh sperm, no significant growth difference was observed between two groups of fry which indicated the possibility of use of cryopreservation techniques in the commercial fish production.

Objectives:

The objectives of the proposed research project is to improve the quality seed production system of Indian major carps by using cryopreserved spermatozoa.

Methodology:

The methods that will be used in the project to achieve the above mentioned outputs are described below:

a) Procurement of milt, quality assessment and counting of sperm.

b) Dilution and preparation of milt for cooling.

c) Freezing of milt and thawing.

d) Fertilization of eggs

e) Dissemination of cryopreservation techniques among hatchery operators

f) Establishment of cryogenic gene bank
