Study on Inter-specific Cross-compatibility Between T. Dioica and T. Anguina

Abstract/ Executive Summary:

Inter-specific crosses between Trichosanthes dioica (pointed gourd) and T. anguina (snake gourd) were studied at IPSA experimental farm during the kharif season in 1995. Three hundred crosses and 100 selfing were made between and within the species and each procedure was repeated three times. Fruit set did not occur in the inter-specific crosses, whereas 100 percent fruit setting was observed in selfed flower. The causes of failure of fruit-setting might be due to the differences of male and female floral morphology and pollen size between the two species. Other probable causes of cross-incompatibility might be due to the poor pollen germination, termination of pollen tube growth in the styler region, swollen or wrinkled pollen tube in the style, failure of the pollen tube to reach the embryo sac and abortion of ovary through the secretion of toxic chemical substances.

Objectives:

To study inter-specific crossing between T. dioica and its close relative T. anguina (snake gourd).

Methodology:

The materials, Trichosanthes dioica (pointed gourd) and T. anguina (snake gourd) were collected from the Department of Genetics and Plant Breeding, Institute of Postgraduate studies in Agriculture (IPSA), Salna, Gazipur, Bangladesh. The seedlings of both the species (raised from vines of pointed gourd and seeds of snake gourd) were sown on 20 February 1995 at IPSA farm. From seedling to the end of the flowering stage, all intercultural operations were performed as per recommendations. During the experimental period, prophylactic measures against insect pests and diseases were done as and when necessary. Five disease-free healthy plants from each species were selected for crossing. The available female flowers of each species were bagged individually one day before anthesis with a polyethylene bag. In the following morning, the bagged female flowers of pointed gourd were pollinated with the pollen grains of snake gourd and vice versa. Selfing was done in each species simultaneously. Three hundred crosses including reciprocals were done manually for inter-specific crossing and each procedure was reported three times. Within the species, 100 selfings were made and it was repeated for three times. Number of fruit setting among the selfed and crossed flowers were recorded. Male and female floral morphology of the two species were observed from the five randomly selected freshly bloomed flowers based on the size and shape of peduncle, calyx, petal, stigma, style, and ovary. Pollen grain variability of both the species was tasted by using acetocarmine as a stain. Sizes of at least 30 pollen grains were measured with the help of a micrometer under a compound microscope. To observe pollen germination, pollen-adhered stigma, pollen tube growth behaviour in the stigma of the crossed and selfed flower and stigma along with style and ovary were fixed and preserved according to standard procedure. Fruit set did not occur in the inter-specific crosses, whereas 100 percent fruit setting was observed in selfed flower.
