Development of Salt Tolerance Brassica Campestris Lines Through Somaclonal Variation Under Saline Stress

Abstract/ Executive Summary:

To achieve the objectives two sets of experiments were conducted during the period from November 2008 to October 2009 at the Tissue Culture Laboratory at the Department of Genetics and Plant Breeding, Bangladesh Agricultural University Mymensingh. In vitro regeneration ability of five selected Brassica campestris genotypes were investigated along with moderate saline stress. Seeds of these genotypes were germinated in Murashige and Skoog (MS) media containing 0, 3, 6 and 12 dsm^-1 NaCl + KCl solution in a ratio of 1:1. To investigate the effect of different treatments, data on various parameters were collected and analyzed by using stem segment as explants. Highest callus (100%) was found in BARI sarisha-12, BARI sarisha-15 & M-27-E₂-1-5 from stem segment. Among all the treatment, the highest callusing was in MS + 2mg/L BAP + 1mg/L 2,4-D + 2mg/L AgNO₃ (T₂) and lowest (11.11% and 44.44%) in MS medium (T₀). From the study it was observed that shoot regeneration was found to be different due to explants, genotypes and hormone concentration. Among all the concentrations, 2.13 shoot in MS+1mg/L BAP+ 0.5mg/L NAA + 2mg/L AgNO₃ and 1.27 shoot in MS+1mg/L BAP+ 1mg/L NAA + 2mg/L AgNO₃ was found to be the best. The highest root initiation percentage was 66.67% in MS medium supplemented with 2mg/L BAP, 0.5 mg/L NAA and 2mg/L AgNO₃ and in MS medium supplemented with 2mg/L BAP, 1mg/L NAA and 2mg/L AgNO₃ from stem segment of the genotype BARI sarisha-9, BARI sarisha-12 and M-27-E₂-1-5. The genotype Kalyania showed the lowest performance (8.33% respectively).The MS medium supplemented with 2mg/L BAP,1.5 L NAA 2mg/L AgNO₃ and 6ds/ mol NaCl +KCl salt solution showed lowest root induction percentage for all the genotypes. The plantlets with sufficient roots thus obtained were transferred successfully to plastic pots and subsequently to the field. The genotype M-27-E₂-1-5 and BARI sarisha-12 survived in growth room as well as in the field but Kalyania was very poor in survivability both in growth room as well as in the field.

Objectives:

The specific objectives were:

i. To screen of rapeseed genotypes for saline tolerance at seedling stage.

ii. To observe variation among the genotypes for their callus induction ability under different concentrations of NaCl and KCl.

iii. To observe plant regeneration ability of the genotypes under different concentrations of NaCl and KCl.

iv. To establishment of a suitable and reproducible protocol for in vitro regeneration of B. campestris plantlets.

Methodology:

The experiment was conducted at the Tissue Culture Laboratory, Department of Genetics and Plant Breeding, Bangladesh Agricultural University, Mymensingh during the period from November 2008 to October 2009. Five higher performed genotypes based on in vitro screening were selected for regeneration experiment. such as BARI sarisha-9, BARI sarisha-12 BARI sarisha-15, Kalyania , M-27-E₂-1-5. Four culture media were used in the study.

Achievement/Findings:

The study revealed that BARI sarisha-9, M-27-E₂-1-5 and BARI sarisha-12 can withstand salinity to some extent. These genotypes can be brought under breeding program as potential parents for hybridization to produce salt tolerant genotypes. Further investigation using marker assisted selection technique and molecular markers like SSR markers can be very helpful in this regard.
