Influence of Inoculum Size and Different Components of Mymp Medium on the Growth of Saccharomyces Cerevisiae Strain Sc-4

Abstract/ Executive Summary:

A potential strain of Saccharomyces cerevisae (SC-4) was assess for its biomass production in MYMP medium as affected by some factors like inoculum size, biotin and peptone. It was found that maximum cultivation period for yeast strain SC-4 was 18th. By increasing inoculum size (1.0gl^-1) the cultivation period was reduced to 12h. Different concentrations of peptone influenced the growth of yeast strain SC-4. Maximum cell mass was produced with 1.5% and 5% peptone. Cultivation with biotin promoted cell yield from 9.0 gl^-1 10.0gl^-1.

Objectives:

To identify the Influence of Inoculum Size and Different Components of MYMP Medium on the Growth of Saccharomyces Cerevisiae Strain SC-4

Methodology:

Microorganism- Some selected Saccharomyces cerevisiae strains were assesed for their biomass production in glucose and molasses media. It was found that the yeast strains SC-4 produced that highest amount of biomass (9.gl-1) at 300C in MYMP medium containing 12.5% molasses after 18h of incubation. Futher world as done with this strain. Media for cell mass propagation: MYMP : This medium is composed of malt extract (0.3%), yeast extract (0.3%), mosasses (12.5% containing 40% sugar) and peptone (0.5%). Determination of protein content: The Protein content of the samples was determined.

Achievement/Findings:

It was observed that maximum cell mass of 9.0gl^-1 was produced in MYMP medium containing 12.5% molasses with the utilization of 63% of the total sugar after 18h of incubation. Increase in cellular protein was also consistent with cell growth. It was observed that MYMP medium supplemented with biotin in the medium substantiated yeast growth to a great extent. Use of biotin increase cell mass production from 9.0ml^-1. The effect of inoculum size on the production of yeast strain SC-4 in MYMP medium at 30⁰C and pH 5.0. Cell mass of 9.0gl^-1 and 10.0gl-1 were produced within 18h of incubation with inoculum concentration of 0.01 nad 0.5gl-1, respectively. on the other hand, cell mass of 9.0gl^-1 was found within 12h of incubation with the inoculum size of 1.0gl^-1. To observe the effect of different concentration of peptone on cell mass production molasses. Effect of peptone concentrations on cell mass production of strain SC-4 . Highest cell mass was observed with 1.5% peptone. Cell mass production was more or less same with 0.5% and 5% peptone after 18h of incubation.
