In Vitro Node Culture of Potato (solanum Tuberosum L.)

Abstract/ Executive Summary:

Five potato varieties. namely, Cardinal. Patrones. Jhaubilati. Shilbilati and Dohazarilal were propagated in vitro by nodal cuttings from well grown in vitro stock materials on MS media supplemented with 3% sucrose, 0.25%mg/l GA₃ and 2.0 mg/I calcium pathogenic acid. Sucrose of four different sources were used as energy source for the propagules. The varieties did not differ much in their response to growth in vitro and they could be placed in order of regeneration potential: Patrones, Cardinal, Jbaubilari, Shilbilati and Dohazarilal. Indigenous varieties showed more branching tendency than the exotic ones. The plantlets grew rapidly and vigorously in the sucrose of BDH's source. Market sugar was also quite good for regeneration of potato plantlets in vitro.

Objectives:

To find out a potato cultivar with good regenerative activity and the possibility of using market sugar for micropropagation of potato in vitro via nodal cuttings.

Methodology:

The study was carried out at the Potato Research Centre, BARI, Joydebpur with two exotic, Cardinal and Patrones, and three indigenous, Jhaubilati, Shilbilati and Dohazarilal varieties of potato. Sucrose of four different sources, three of them are analytical grade of a) E. Merck, West Germeny, b) BDH, England, C) CDH, India and the rest market sugar were used in this study. The purity percentages of sucrose of different sources are as follows: E. Merck-99.91%, BDH-99.92, COH-99.88% (purity percentages noted from the lebel of the bottles) and market sugar-98.78% (personal communication). The basic cuIture media consisted of agar-0.8%, the inorganic and organic constituents of Murashige and Skoog and 3% sucrose with 0.25 mg^-1giberrellic acid and 2.0 mg-¹ calcium pantothenic acid. The pH of the media was adjusted to 5.6 prior to autoclaving at 121°C for 20 minutes at a steam pressure of 15 psi. The stock plants were obtained through meristem culture on the media described earlier. The isolated meristems were cultured at 22°-24°C and 16 hrs. illumination of 2000-3000 lux daily. Agar was used at the rate of 0.6% instead of 0.8%. Plantlets were in vitro multiplied for necessary nodal culture. Stem segments, from forty days old plantlets, containing one auxiliary bud and its subtending little leat, were cut and put onto the nutrient media in 125 X , 5mm test tub e s. The tubes were sealed with heavy duty aluminum foil. All these operations were done aseptically and the cultures were incubated in conditions similar to that of excised meristem culture. Auxiliary buds were used for direct development of plantlets. The explanst that have died because of contamination or injury were not considered for the evaluation. The vigour of the plantlets was measured visually. Data on percentage of successful cultures, days to new shooting and rooting, height, number of leaves, roots and branches per plantlet, vigour end rate of growth (mm/dav) were recorded.
