In Vitro Propagation of Gerbera (gerbera Jamesonii)

Abstract/ Executive Summary:

The experiment was conducted at Sher e Bangla Agricultural University during October 2013 to August 2014 to establishment of in vitro plant regeneration protocol in Gerbera and Micro propagation of Gerbera. The explants of shoot tips, immature inflorescences, capitulum explants, axillary buds, leaf with mid-rib and leaf blade segments as well as flower stalks obtained from selected varieties of gerbera (Gerbera jamesonni Bolus.) cultivated in Bangladesh were exploited for callus induction and in vitro regeneration of plantlets. Among the explants flower stalk were suitable and superior for callus induction. However, highest numbers of multiple shoots were obtained when the flower bud derived callus was subcultured on MS supplemented with 4.0 mg//I BAP and 1.0 mg/INAA. Root induction at the base of the in vitro regenerated shoots was tried using MS containing different concentrations of IBA. Best root induction was observed MS supplemented with 0.3 mg/I IBA.

Objectives:

Methodology:

The experiment was conducted at Sher e Bangla Agricultural University during October 2013 to August 2014 to Establishment of in vitro plant regeneration protocol in Gerbera and Micro propagation of Gerbera.

Source Material

Plants of gerbera (Gerbera Jamesonii Bolus.) varieties having red, yellow, orange, pink, golden and white coloured petals collected form Jessor and were used for this study. Tender leaves and 2.5 – 4.0 cm long flower buds (9-11 days old) along with flower stalks were collected from field grown materials.

Shoot formation:

In order to evaluate the shoot formation ability, the produced calli were cultured on different shoot formations media.

Root formation

The produced shoots were transferred to the rooting medium containing different combination and incubated for 3-4 weeks.

Achievement/Findings:
