Detection of Mycobacterial Infection from the Lymphnode and Lungs of Suspected Cattle Using Traditional and Molecular Technique

Abstract/ Executive Summary:

A total number of 14 samples for the present study were collected from the cattle of Milk Vita Sirajgonj, CCBDF (Central Cattle Breeding and Dairy Farm), Savar, Dhaka; Military dairy farm, Savar, Dhaka; CDIL (Central Disease Investigation Laboratory), Dhaka; Kawatkhali, Masuabazar and Muktagacha slaughter houses of Mymensingh District during the period of January-June, 2011. Prescapular and mesenteric lymphnode, lung and blood were collected. Routine analysis of blood was carried out but did not reveal any significant change. Samples (Portion of lymphnodes and other tissues like lung) for molecular analysis were preserved at 20? and samples for histopathology (portion of lymphnodes and other tissues) were collected in 10% buffered formalin. Collected samples were subjected to H & E staining, acid fast staining and DNA extraction for PCR amplification. Acid fast staining of lymphnode smears successfully detected Mycobacterium in two out of 14 samples. Polymerase Chain Reaction (PCR) was done with the extracted DNA, Mycobacterium complex specific primers were used and three samples were found positive. This study suggests that the PCR technique is a useful and rapid diagnostic test for the identification of Mycobacterium complex in dairy cattle. PCR takes only a few hours to detect Tuberculosis in dairy cattle with a high degree of sensitivity and specificity.

Objectives:

To determine the detection of mycobacterial infection from the lymphnode and lungs of suspected cattle using traditional and molecular technique.

Methodology:

The research work was performed in the Department of Pathology, Faculty of Veterinary Science, Bangladesh Agricultural University, Mymensingh. A total number of 14 (fourteen) samples for the present study were collected from the cattle of Milk Vita Sirajgonj, CCBDF, Savar, Dhaka; Military dairy farm, Savar, Dhaka; CDIL, Dhaka; Kawatkhali, Masuabazar and Muktagacha slaughter houses of Mymensingh during the period of January-June, 2011. Among the 14 samples four samples are taken from live animal and ten samples are taken from slaughtered cattle. Whole blood and superficial lymphnode biopsy were collected from the suspected live animals. However, to ensure positive reactivity, following necropsy of the animal, mesenteric lymphnode and other tissues or organs including lungs were collected. All samples were stored as required until use. Blood slides were stained with Modified Wright’s stain, examined under oil-immersion objective and 100 cells of WBC were counted. The blood smear was fixed by absolute methanol and stained with Giemsa stain for 30-45 minutes. After washing, slides were air dried and examined under oil-immersion objective and 100 cells of WBC was counted. The WBC pipette was filled with the diluting fluid (1% HCl) upto 11 marks and shaked for 2-3 minutes by moving it with right hand in the way of 8 knot motion for mixing the blood and diluting fluid. 1-2 drops of mixture was discarded from the pipette and a small drop of it was placed on one side of the chamber and the cover slip was touched to spread the mixture on the counting chamber. All the 4 square chambers of WBC from the 4 corners were counted and recorded.

Achievement/Findings:

Tuberculosis in dairy animal is gradually increasing in Bangladesh. Mycobacterial infection is commonly seen in the establish dairy farm in Bangladesh. Tuberculin test is used for the detection of mycobacterial infection in animal. PCR is a technology that requires a minute amount of bacterial DNA and species specific reaction can be generated. Tuberculin test positive animal can be used in PCR evaluation of species specific reaction. An understanding of mycobacterial species specific reaction would enable us to design future strategy to prevent further dissemination and designing a control strategy.
