Screening of Simple Sequence Repeat (ssr) Marker for Polymorphism in Maize

Abstract/ Executive Summary:

Maize inbred lines obtained from CIMMYT and Inbred lines developed by BARI, using SSR markers. DNA polymorphisms were detected by separation in agarose gel. A total of 25 informative SSR loci were chosen for initial screening of eight inbreds. Amplification products were separated by electrophoresis in a horizontal gel system.

Objectives:

To investigate the level of genetic diversity among maize inbred lines using SSR Marker.

Methodology:

Twenty seven maize inbred lines and hybrids were used in the experiment. DNA was extract following CTAB method. The SSR primers used in the current study were obtained and chosen based on repeat units and bin locations to provide uniform coverage of the entire maize genome. A total of 25 informative SSR loci were chosen for initial screening of eight inbreds. Amplification products were separated by electrophoresis in a horizontal gel system at 100V for 2.0 h on 0.8 % agarose gels (stained with ethidium bromide (0.5 mg ml_1) using 0.5_/ TBE buffer (89 mM Tris, 89 mM borate, 2 mM EDTA pH 8.3). Finally, the gels were photographed under UV light (Geldoc Bio-Rad) attached to a gel documentation system with Biocapture software (Vilber Lourmat, France). After completion, microplate was taking out and stores it in –20°C or at 4°C with dye.

Achievement/Findings:

Initially only three SSR primer, were screened. Out of three one showed polymorphic band among the inbred lines.
