Reproductive Potential of Bombyx Mori L. Induced by N-nitroso-n-ethylurea in Egg Stage

Abstract/ Executive Summary:

The effect of the mutagen, N-nitroso-N-ethylurea (NEU) as an enhancer of fecundity was tested on four varieties of the silkworm Bornbyx moni L. viz . Nistari-M . Nistani - G. RB-i 12 and BSRI-801 at concentrations O(control). 20, 25, 30 mM . N- nitroso-N-ethylurea at 20 mM was most effective in egg production for all the parental lines and their successive crosses . while 30 mM was effective to produce the lowest number of eggs . Similar results were obtained for egg-viability. Genotypes of the silkworm and the concentrations of the mutagen used had significant effects on the fecundity of B. mori.

Objectives:

To find out the effect of the mutagen, N-nitroso-N-ethylurea (NEU) on the reproductive potential of four indigenous varietes of B. mori, viz. Nistari-M, Nistari-G, RB-i 12 and BSRI-80 1 under different crossing schedules.

Methodology:

The eggs of four varieties of B. mori viz ., Nistari-M, Nistari-G, RB-i 12 and BSRI-801 were collected from the Germplasm Bank of Bangladesh Sericultural Research and Training Institute (BSRTI). Rajshahi. Nistari -M was isolated from the original indigenous Nistari variety and Nistari-G, was known as a poor silk yielder developed by hybridization. RB- 112 is a bivoltine variety which was developed through a selection programme from the exotic stock of K.A. Pure Mysore, exhibiting good performance during pre-winter season. BSRl-80 1 is a high yielding multivoltine variety developed at the BSRTI by crossing Nistari-c/ 43 ( 1) and MS4. Four of NEU concentrations, e.g. O(control), 20, 25 and 30 mM (millimol) were prepared by adding requisite amounts of the mutagen in distilled water. The concentrations were placed separately in an air-tight desiccator and were left for 12 hr at 25^oC. The mutagen was induced in eggs after 24 hr of their opposition. Three replicates were made in each of the NEU treatments. An untreated control was also maintained in three replicates throughout the experiment. The experiment was conducted using one parental lines and their F 1 hybrid populations. Silkworm larvae were reared according to the methods of Krishnaswami ( 1978). The following cross schedules were employed : a : Ntstari-M, b:Nistari-G, c: RB-112, d: BSRI-810 and their crosses: e: axb, f: bxa, g: axc, h: cxa, i: axd, j: dxa. k: bxc, l: cxb. m: bxd . n: dxb . o: cxd and p: dxc. The total number of eggs laid by an individual female gave the fecundity. The total number of dead and unfertilized eggs constituted unhatched egg. The experiment was carried out at 25±1oc and 80±5% RH .

Achievement/Findings:

In considering the hatching percentage the results showed a high level of significance (P< 0.001) in crosses and concentrations excepting their interactions. The maximum hatchability was recorded for 25 mM in almost all the crosses. From the present study it was clear that treatments with 20 performance in terms of and 25 mM NEU showed an acceptable higher reproductive potential. Note that NEU which is a mono-functional alkylating super-mutagen, was capable of poroducing a well defined spectrum of ethyl attacks in the DNA of treated cells. The effects of the mutagen on D. melanogasterwere discussed. The percentage of dominant lethal induction was directly dependent on the mutagen concentration . The results from this and previous studies on reproduction indicate that NEU can drastically change the reproductive potential of F 1 silkworms .
