Graft Union Studies in Mango

Abstract/ Executive Summary:

Transverse sections of the successful graft union showed that parenchymatous cells proliferated from both the stock and scion. It was further observed that the thin walled cells of xylem rays in most recently formed xylem, cambial layers and most recently formed phloem rays were involved in the production of the callus tissues. The vascular meristem appeared first where the cambium of the stock and the scion were ill contact with the callus tissues. Differentiation of the normal secondary xylem and secondary phloem from cambial layers in callus was observed.

Objectives:

To determine the graft union studies in mango

Methodology:

One hundred seedlings of local mango varieties, growing for about a year in the nursery of the Bangladesh Agricultural University Horticultural Farm, were used as rootstocks. In the month of March 1972, mulching operation was done to these seedlings and a teaspoonsful of urea was applied to each of the seedlings followed by thorough irrigation. When the seedlings started to produce new leaves and when the bark of the seedlings could be pulled out easily, they were taken to be ready for grafting operation. Terminal shoots were selected from Fazli mango that grew in the previous season. One week before the grafting operation, all the leaf blades in each of them were trimmed off leaving their petioles intact (3). Fifty venner and fifty side grafts were prepared utilizing the pre-treated terminal shoots. Grafting operations were done in the 1st week of May, 1972 For anatomical studies of the graft union, a good many samples of both venner and side grafts were collected after two months from the date of operation. The samples, being woody in nature, were cut into pieces of I em length and were fixed in FAA solution (70% ethyl alcohol 90ml, glacial acetic acid 5ml, 40% formalin 5ml). The samples were taken out from the fixative and washed in running water for 48 hours. The woody samples were softened by treating them with hydrofluoric acid (diluted with twice volume of water) for two weeks (4). The treated samples were thoroughly washed in running tap water for 48 hours. The cut pieces were then dehydrated an embedded in paraffin. Transverse sections ( 10-15 micron thick) were cut with sliding microtome. Sections were stain with safranin-fast green and mounted on Canada balsam.

On examination of transverse section cut through the successful graft union of both venner and side grafts, it was observed that parenchymatous cells (callus) were proliferated from both the stock and scion. From these observations it might be concluded that one of the prerequisite for successful graft union should be that the cut surfaces of the various tissues especially the cambial layers should be brought into close proximity as far as possible at the time of making graft union.
