In Vitro Clonal Propagation of Guava (psidium Guajava L.)

Abstract/ Executive Summary:

Nodal explant was found better than shoot tip explant for multiple shoot regeneration in MS medium supplemented with 1.0 mg/I BAP. Kinetin was found less effective for shoot proliferation than BAP. The regenerated multiple shoots were devoid of any root. The best rooting was observed when the isolated individual shoot was cultured one half strength MS medium supplemented with 0.5 mg/I ISA. Rooted plantlets from the culture jars were transferred to a mist house and then successfully established in the soil.

Objectives:

To establish an efficient and reproducible in vitro propagation technique for a popular guava variety, 'Kazi piara'

Methodology:

Preparation of explants: The plant materials were collected from 3-4 months old seedlings grown in a greenhouse. Shoot pieces of 3-4 cm long were taken and then placed in a casserole dish with 7 per cent domestos (Sodium hypochloride) for 25 minutes and then washed 4 times with sterile distilled water. The shoot pieces were then aseptically cut into about 0.75 cm ^{- 1} cm long pieces. The basic culture medium was composed of MS salts supplemented with 3 per cent sucrose and different concentrations (0, 0.1, 0.5, 1.0, 5.0 mg/I) of BAP (Benzylaminopurine) and Kin (Kinetin), respectively. The medium was solidified with 0.8 per cent Difco Bacto agar. The medium was adjusted to pH 5.8 and then sterilized in an autoclave at 121°C and 15 psi for 25 minutes. Single explant was incubated in a 30 ml jar containing 10 ml medium to a depth of 2-3 mm and maintained at 25 ± 2°C under fluorescent light (1000 ) following 16h day (light) : 8h night (dark) cycle. Shoot initiation started after 7-12 days incubation of the culture. For sub-culture, every 3-4 weeks after, the explants were trimmed under aseptic conditions by cutting the basal end and finally incubated into the fresh medium with the same concentrations used before. Fifteen observations were taken from each treatment and repeated twice. Data on shoot number and shoot height (cm) were taken after 8 weeks of culture. When individual axillary shoots (initiated from the explants) grew upto 3-4 cm, were excised and transferred to a modified MS medium supplemented with different concentrations (0, 0.05, 0.1,0.5,1) of IBA (Indole-3-butyric acid). Root initiation started within 7-15 days of transfer. Fifteen shoots were taken for each treatment and repeated twice. Data on root number and root length were taken after 5 weeks of culture. Rooted plantets thus obtained were taken out of the culture jars, washed thoroughly to remove the medium and planted in small plastic pots containing soil. During the first 7- 10 days, the potted plantlets were kept in a mist propagating house for establishment.

Rooted plantlets from the culture jars were transferred to a mist house and then successfully established in the soil.Nodes are capable of producing multiple shoots in vitro and subsequently these shoots can be rooted to develop complete plants.
