Reaction of Some Betel Vine Lines to Leaf Spot (colletotrichum Cepsict)

Abstract/ Executive Summary:

An experiment was conducted at the Regional Agricultural Research Station, Rahmatpur, Barisal for three consecutive years to screen betel vine lines against leaf spot (Colletotrichum capsici). Inoculation with spore suspension was made and the lines were scored with 0-3 rating scale. The line BV010 (Sanchi) was found resistant. Among the other lines, BV005 (Chaltagota) and BV007 (Gachpan) were moderately resistant while BVOOl (Banglapan), BV002 (Bawla), BV003 (Bhabna), BV006 (Chherfuli), BV008 (Mithapan), BV009 (Mohanoli), BVOll (Zyloo) were moderately susceptible. BV004 (Bhola) showed susceptible reaction to the pathogen.

Objectives:

To screen betel vine lines against leaf spot (Colletotrichum capsict).

Methodology:

The experiment was conducted at the Regional Agricultural Research Station, Rahmatpur, Barisal during 1989-90 to 1991-92. There were eleven betel vine lines in the experiment viz., BV005 (Chaltagota), BV006 (Chherfuli), BV007 (Gachpan), BV009 (Mohanoli) from Barisal; BV001 (Banglapan) from Rajshahi; BV002 (Bawla) from Mymensingh; BV003 (Bhabna) from Jessore; BV004 (Bhola) from Bhola; BV008 (Mithapan) from Narail; BV010 (Sanchi) from Nawabgonj and BV011 (Zyloo) from Bagerhat. Cuttings were taken from six month old betel vine which were planted on raised beds in double row system. Each bed was 2.5 m long, 30 cm wide and 15 cm high. Adjacent beds were seperated by 50 cm wide drain. Cuttings were planted 10 cm apart in line. Mustard oil cake was applied @ 6 ton/ha/annum in equal installments twice monthly starting from June to November. The experiment was set up in RCB design with three replications in a newly constructed Boroj. Intercultural operations, such as weeding, prunning, tieing, lowering of vines etc. were done as needed. Plants were inoculated by spraying with spore supension at two months plant age. For preparation of inoculum, infected leaves were collected from farmer's fields near the Regional Agricultural Research Station, Rahmatpur, Barisal. Pure isolation of pathogen was made from the infected leaves and cultured on PDA medium. Fourteen days old culture was blended in sterilized water. The spore suspension was then filtered with a fine cheese cloth. Then the suspension was diluted to 10⁶ spores/ml. Disease incidence data were recorded three times per year first after two months of inoculation following 0-3 rating scale.
