Screening of Eggplant Germplasm Against Bacterial Wilt in Bangladesh

Abstract/ Executive Summary:

Twenty three local and exotic cultivars of eggplant and three wild relatives of the crop were screened in the glass house against Bacterial wiIt (Pseudomonas solanacearum E. F. Sm) during September-October, 1980 at Citrus and Vegetable Seed Research Centre, BARI, Joydebpur. The wild relative Solanum mammosum was found resistant while the cultivar Dingaras Multiple Purple and Khatkhatia Long were found moderately resistant. The cultivars Nayankazal, Khatkhatia round, Taiwan naga, Nagasaki naga and Kurume naga were found moderately susceptible whiIe the rest of the cultlvars as highly susceptible.

Objectives:

To evaluate the tolerance of the cultivar and its relatives against bacterial wilt.

Methodology:

The experiment was conducted in the glasshouse during September-October. 1980. The study included thirteen local and ten exotic cultivars of eggplants and also three wild relatives of the crop. Seeds were first germinated in incubator and then sown in polythelene pots filled with sterlized soil. The experiment was laid out in RCB design with three replications. Thirty plants of each cultivar were tested for the study. Pure culture of Pseudomonas solanacearum E. F. Sm. was prepared on Potato Dextrose Agar medium by isolating the organism from the eggplant showing typical symptoms of bacterial wilt. The identification of the isolate was confirmed by Cock's Postulate. Root dipping method was followed for inoculation. Five weeks old seedlings were uprooted and one-fourth root was cut by sterilized scissors. Then the roots of each seedling were dipped in bacterial suspension for 5 minutes and were replanted in the same pots. The inoculated seedlings were observed daily for disease development. Data were collected 7,9 and 11 days after inoculation. The degree of disease development of each cultivar was expressed by using Disease Index (D. I.). and percent of plant infected (P.I.). The disease severity index was calculated by using the following formula. Disease Index= 1 x n₁ +2x n₂+3x n₃ / n₀+n₁ + n₂+n₃ Where 0 stands for no infection, 1 for slightly wilting, 2 for rather wilting, 3 for severe wilting and n stands for number of seedling. Cultivars having D. I. upto 0.50, 0.51 to 0.99, 1.0 to 1.50 and above 1.51 were considered as resistant, moderately resistant moderately susceptible and susceptible respectively.
