Host Status of Some Crops to Meloidogyne Incognita

Abstract/ Executive Summary:

Seventeen crops were grown in soil infested with root-knot nematode (Meloidogyne incognita) to determine their host status on the basis of the presence at root-knots, larvae and mature females in roots. Nine crops were graded as excellent hosts, five as poor and three as non-hosts.

Objectives:

To determine the host status of some crops to meloidogyne incognita

Methodology:

Sufficient inocula were built up from egg-maas cultures of M. incognita on tomato and brinjal grown in nematode free soil. The species was confirmed by the host differential test developed by sasser. The soil was spread in a layer 2cm thick on a plastic sheet and exposed it to the sun for 8 days to eliminate the nematodes if any present. Inoculum consisted of mature egg masses, root- knots and larval suspension. Large quantities of inoculum were added to the soil and mixed thoroughly to ensure uniform distribution. One wooden box measuring 2.5m long, 1m wide and 0.34m deep was filled with the infested soil. Fifteen seeds of each crop were sown in the box in 17 lines each representing a treatment. After seven days of germination, 5 seedlings were removed and 10 seedlings line were grown. After 45 days, 5 plants from each line were carefully uprooted, washed and the knots were counted. To assess the number of nematodes within the roots, the roots of the remaining plants were similarly cleared, chopped off and mixed thoroughly. One gram root from each sample was stained with cotton blue lactophenol for 3 minutes and distained with clear lactophenol for 48 hours and the number of larvae and mature females present in the roots were counted under sterioscopic microscope.

Achievement/Findings:

The non-hosts and the poor hosts identified in the present investigation may profitably be used in developing crop rotation sequences to combat root-knot nematode, M. incognita.
