Chemical Seed Priming Influence on Germination and Vigour of Rice Seedling

Abstract/ Executive Summary:

An experiment was conducted at the Seed Laboratory, Department of Agronomy, Bangladesh Agricultural University, Mymensingh during the period from September to December 2008 to study the effect of chemical priming of rice seed (cv. BRRI dhan 29) on their seedling germination and growth. Rice seeds were soaked respectively in 1%, 2% and 3% solutions of KCl, CaCl₂, and NaCl and also in water for 15, 20 and 25 hours. The experiment was laid out in a randomized complete block design (RCBD) with three replications. Germination and vigour tests of the seed under treatments were done in sand culture at moisture level of 60 % of field capacity. Vigour parameters such as mean germination time, vigor index, shoot length, shoot dry mass, root length and root dry mass of rice seedling were measured in the trial. The results revealed that priming treatments had significant influence on the germination and all the growth parameters of rice seedlings. Priming with 2% KCl for 25 hours showed the highest germination and the lowest mean germination time. Priming with 3% NaCl for 15 hours produced the longest shoot length and the highest shoot dry mass. Priming with 1% KCl for 15 hours showed the highest root length while 3% KCl for 25 hours showed the highest root dry mass. The seed having no priming treatment showed the lowest germination, vigor index, shoot length, shoot dry mass, root length and root dry mass and the highest mean germination time. The present study concludes that priming of rice seed by soaking in 2% KCl solution for 25 hours could improve germination and growth of rice seedling.

Objectives:

To study the effect of chemical priming of rice seed on germination, vigor and early growth of seedling.

Methodology:

The experiment was conducted at the Seed Laboratory of the Department of Agronomy, Bangladesh Agricultural University (BAU), Mymensingh during the period from September to December, 2008. Rice seed of variety BRRI dhan29 was used as the test material in the experiment. With distilled water 1%, 2% and 3% solutions of KCl, CaCl₂, and NaCl were prepared and the seeds were soaked for in the solutions and also in water for 15, 20 and 25 hours, respectively. A non treated control sample was also used in the trial. The experiment was laid out in a Completely Randomized Design (CRD) with three replications. Germination and vigour tests were conducted on sand culture method using sand media at 60% of its saturation. The data on germination were recorded by daily counting of germinated seed from the first day after sowing and continued up to 14 days. Mean germination time and vigor index were calculated from the daily recorded data. On the 14th day of setting the test the numbers of normal seedlings were counted and the germination percentage was recorded. Length of root and shoot of seedlings were recorded. Randomly selected 10 normal seedlings from each Petridis was taken to record data on shoot and root length. The seedlings after the measurement of shoot and root length were separated and then dried in an electric oven at 72^oC for 48 hours. After drying the dry weight of shoot and root were recorded by an electric balance. The collected data were compiled and analyzed statistically using the analysis of variance (ANOVA) technique and the means were compared by Duncan’s Multiple Range Test.

Achievement/Findings:

The present study concludes that soaking of rice seed with 1%, 2% and 3% solutions of KCl, CaCl₂ and NaCl enhances germination and vigour of rice seedlings. However, the best result could be obtained by soaking of seeds in 2% KCl solution for 25 hours.
