Storage of Cut Flowers by Formation of Intracellular Water Structured Through Applying Xenon Gas

Abstract/ Executive Summary:

Xenon, an inert gas caused to make the water structured into the intracellular water of living cell at a pressure and temperature. This phenomenon was supported b), the appearance of chemical shift obser.vecl by proton nuclear magnetic resonance (NMR) measurement. Finally the method was applied to cut flowers in order to make intracellular water structured, which caused to suppress the respiratory metabolism of living cells. Two groups of cut flowers were placed in high-pressure tight steel container. Xenon gas was superimposed into one high-pressure steel container up to the partial pressure of 0.25 MPa and the other container was filled with air of atmospheric pressure as a control. They were stored for 10 clays at 275K. The xenon partial. Pressure reduced due to its dissolution into intracellular water of cell and it became finally 0.223 MPa. After 10-day storage, both two containers were ventilated to avoid the injury by high CO₂ atmosphere and both groups of cut flowers were stored again as the 2nd phase of storage at the same temperature in the atmospherics air. The residual effect of xenon was clearly understood from the test results that the shelf life of cut flowers was extended upto 16 days of storage compared to the non-treated or controls one. From the study it may be concluded that, Xe could be used for the safe storage of high valued horticultural crops or ornamental plants.

Objectives:

To determine the Storage of Cut Flowers by Formation of Intracellular Water Structured Through Applying Xenon Gas

Methodology:

Xenon gas was selected in this study for making intracellular water structured of cut flowers because it is the most soluble gas among the inert gases. When, xenon dissolved into water, structured water was formed. In order to confirm this phenomenon experimentally, Nuclear magnetic resonance (NMR) measurement was conducted to observe the appearance of chemical sift. Two Pressure tight sample tubes similar inner dia (10mm) and log (180 mm), were filled with distilled water to the height of 40 mm. After degassing of both sample tubes, xenon was superimposed to one sample tube in order to prepare the structured water. The other sample tube with degassed distilled water was used as a control. Proton NMR spectrum of each sample was observed by JOEL JNM-A 500 FT-NMR system. Cut flowers of Nasu polo, a Japanese cultivar showing almost same growing stages were collected from the local flower market of Tokyo, Japan. Immediately after harvest this flower, the flower traders stored the cut flowers in a controlled condition for one day. Immediately after collection from the traders, the flowers were kept in a thermal insulated bag and within 1-2 h. and stored in the controlled room temperature for 1 day at 274 K before onset of the experiment. The samples were placed inside the high pressure steel container (2.123 x 10^-3m3) having inside diameter of 130 x 10^-3 m and height of 160 x 10^-3 m. The experiments were set up for 16 days. IN each experiment, two containers were used where one container was treated as control i. e. without applying any pressure, Pressure was maintained for 10 days, and on the 11th day, the gas was ventilated gently by releasing the pressure gradually.

Achievement/Findings:

Proton NMR measurement indicated the formation of structured water when xenon gas dissolved into distilled water. Metabolic activity was suppressed in the xenon treated cut flowers. As a result freshness maintained and extended the shelf life of cut flowers upto 16 days. From the study it may be concluded that, Xe can be used for the storage of high valued ornamental plants or horticultural crops.
