Application of Biometric and Electrophoretic Methods for the Stock Discrimination of Hilsa (tenualosa Ilisha) in Bangladesh Water

Abstract/ Executive Summary:

The results of the discriminant function analysis of both morphometric and meristic characters showed that out of 19 morphometric and 5 meristic characters, 12 morphometric and 3 meristic characters were highly significantly different from each other habitat area (Chandpur. Goalando, Chittagong and Khulna). Hence, there is the possibility of having at least four stocks of hills a in Bangladesh. On the other hand, electrophoretic analysis showed that the allele frequency difference was observed at G3PDH-2. only for Kuakata population. Kuakata belongs to marine environment and other three stations (Goalando. Chandpur and Hatia) belong to riverine and estuarine environments. Therefore, there is a possibility of having two stocks/races of hills a, one the riverine stock and the other marine stock in Bangladesh water.

Objectives:

To investigate the stock/ races of hilsa through both the biometric and electrophoretic methods.

Methodology:

The present study was designed to explore the stock variability, either hilsa comes from a unit stock or from several stocks in Bangladesh water. Under this experiment two methods were applied to verify the hilsa stock/races; one was biometric method and another one was the electrophoretic method. Both the methods are depending upon the genetic variability. The same fishes were not included for both the methods. The sampling places for hilsa population were considered on the basis of different habitat areas. Sampling stations, viz. Chandpur, Goalando, Khulna and Chittagong, which represent the four different habitats of rivers the Meghna, the Padma, the estuary and the sea respectively. From each station at least 25 specimens were collected monthly during the period of November 1987 to October 1989. From each specimen nineteen morphometric characters and five meristic characters were measured and counted for the comparison, either hilsa population were in morphometrically and meristically significantly different from each other habitat or not. The collected data were analyzed by the advanced computer statistical module of the SPSSPC⁺ software package. For the electrophoretic study, about 40 alive young hilsa were collected from each sampling site, specifically samples of eye, liver, heart and abaxial muscle tissue were dissected from each specimen. Tissue samples were placed in labeled cryotubes and preserved in liquid nitrogen. Each type of samples were collected from 4 different habitats, viz. Goalando, Chandpur, Hatia and Kuakata. The tissue samples were stored in a liquid nitrogen container immediately after collection and later on carried out to the lCLARM lab, Philippines. The studies were conducted during the period of December 1992 to July 1993. The starch gel electrophoresis technique was applied for the analysis of the collected samples and Biosys-1 package programme was applied. Single-individual genotypes were recorded as alphabetical characters. The unweighted pair-group method with arithmetic averaging (UPGMA) in the CLUSTER subroutine of Biosys was used to estimate genetic distances for the linkages among species.

Achievement/Findings:

Electrophoretic results showed the possibility of having two stocks of hilsa in Bangladesh water whereas, only one spawning ground of hilsa has been identified at the coastal belt region which do not support the cushing's view. Because, two stocks of hilsa should have two spawning grounds, but practically two spawning grounds have not yet been identified in Bangladesh water which predict that there might have single stock of hilsa population in Bangladesh that is the contradiction to the electrophoretic result. Therefore, it is assumed that it might so happen that marine hilsa as well as riverine hilsa are spawning intermingling at the same (identified) spawning ground. Again, it could happen due to the highly migratory nature of the fish.
