Studies on Semen Characteristics of Different Crossbred Bulls

Abstract/ Executive Summary:

The present study was undertaken to evaluate the effect of genetic group of bull on semen characteristics. Semen was collected from thirty five breeding bulls belonging to six genetic groups (75%Holstein Friesian (HF)x25%Local (L), 50%HFx50%L, 62.5%HFx37.5%L, 87.5%HFx12.5%L, 68.75%HFx31.25%L and 75%Sahiwal (SL) x25%L). Genetic groups had a significant (p<0.05) effect on semen characteristics. The significantly (p<0.05) highest semen volume per ejaculate (cc), % individual motility (progressive) and sperm concentration (million/cc) were found in 87.5%HFxI2.5%L and lowest in 50%HFx50%L. Furthermore a positive correlation between volume of ejaculate, motility and sperm concentration were also found. It was concluded that 87.5%HFx12.5%L genetic group was suitable for most of the semen parameters than other groups.

Objectives:

To evaluate the effect of genetic group of bull on semen characteristics.

Methodology:

The information on semen characteristics of thirty rive breeding bulls was collected on the basis of records maintained at District Artificial Insemination (AI) Centre, Rajshahi during the period from 2001 to 2003. The bulls under study were divided into six groups according to their genetic composition as 75%Holstein Friesian (HF) x 25%Local (L), 50%HFx50%L, 62.5%HFx37.5%L, 87.5%HFx12.5%L, 68.75%HF x 31.25%L and 75%Sahiwal (SL)x25%L. The age of the bulls was determined by inquiring the date of birth from bull register which ranged from 13 to 19 months. Bulls were maintained under identical feeding and management system. Bulls were tested for fertility before putting in breeding herd. Semen was collected early in the morning at 3 to 5 days intervals with the aid of an artificial vagina (A V). The semen collected was brought to the laboratory immediately and was placed in water bath at 37° C for evaluation. Parameters recorded were: volume of ejaculate (cc), motility (%) and sperm concentration (mill./cc). Sperm concentration was measured using an improved Neubaur haemocytometer (Salisbury et al. 1978). Motility, as a percentage 01" individually motile spermatozoa, was estimated by examining a drop of fresh

semen on a warm slide (with cover slip) under a microscope at 40x. Motility was scored on the basis of the percentage of spermatozoa with normal forward progressive movement while those showing circling movements or oscillating at one place were regarded as nonprogressively motile.

Furthermore a positive correlation between volume of ejaculate, motility and sperm concentration were also found. it can be concluded that 87.5%HFx12.5%L genetic group was better for most of the semen characteristics and it can be used for artificial insemination which will further improve the conception rate in cow.
