Peat soil as carrier material for biofertilizer was collected from eight different locations of Bangladesh. Physical and chemical properties of soils varied widely. Survival of groundnut rhizobia on various peat samples at normal temperature upto 45 days has shown that the highest (6.1 X 109) and lowest (1.9 X 109) cells count was obtained with" Gobra" and "Mollahat'' peat respectively. A positive correlation was obtained between physical and chemical characters of peat and viable count of groundnut rhizobia. On the other hand, survival of groundnut rhizobia in peat varied with different temperature. Survival was best at low temperature (4°C ± 1) followed by room temperature (25°C ± 4). Cell number declined rapidly at elevated temperature (370C+ 1). Statistical data show that the effect of temperature, storage duration and their interaction is highly significant.

To Studies on the Survival of Rhizobial Isolates in Indigenous Peats and their Suitability as Carrier Materials of Biofertilizer
 

Peat was collected from eight locations i. e. (i) Tungipara, (ii) Chamta, (iii) Gobra, (iv) Haridashpur, (v) Udaipur, (vi) Mollahat (vii) Narail and (viii) Panchkohania. All the samples were collected between depth of 180-300 cm. Peat samples were air-dried and crushed to a fine powder using a sheat grinding mill and then passed through a 100 mesh seive. The following physical and chemical properties were analysed: (i) maximum water holding capacity; (ii) pH; (iii) total nitrogen; (iv) available P; (v) exchangeable K; (vi) organic carbon; (vii) organic matter; (viii) C/N ratio; (ix) exchangeable Ca. All the collected peat samples were acidic in nature. The samples were ground and neutralized with lime (amount varied with the sources of peat). Two kg of peat was then dispensed in cotton bag and autoclaved at 15 1b pressure for four hours. After sterilization, 100 g peat samples were packed asceptically into polythene bags. Broth cultures (grown in yeast mannitol liquid media) of indigenous groundnut rhizobia (CN- 307) was injected asceptically into the separate polythene bags using a sterilized syringe to bring the moisture content of the carrier material to 40 % by weight. The inoculated materials were incubated in an incubator at 280C (±1) for 7 days. Then incubated. materials were kept at normal (room) temperature upto 45 days. The packets of Gobrapeat were then sub-divided into three parallel sets. One set was stored in the laboratory at room temperature 250C (± 4), the second set was stored in a cooled incubator at 40C (± 1) and the third set was stored in an incubator at 370C (± 1). The viable count of groundnut rhizobia in the peat inoculant was made at 0,15,30,60,90, 120 and 150 days of storage. Counts were determined using the drop plate method.

In Bangladesh, the most practical achievable standard would appear to be 108 per gram peat with a relatively high application rate of 1 g inoculant per 20 g seed, thus assuring 106 rhizobia per seed. In addition, it is recommended that inoculants should be used within 70/80 days of manufacture. Using this recommended inoculant technology, subsequent experiments have given consistent response; both in experimental sites and in farmers field. Inoculation using mixed strains at 108 viable cells per g applied at 1 g/20 g seed gave 20-40% increase in yield of groundnut. It was observed that low temperature was found to be suitable for survival of groundnut rhizobia in peat as compared to room temperature. A positive correlation was obtained between physical and chemical characters of peat and viable count of groundnut rhizobia.
