Screening of Blackgram Genotypes Based on Sulphur and Nitrogen Uptake and Transport

Abstract/ Executive Summary:

Pot culture experiments were conducted to screen high nutrients uptake genotypes of blackgram. The tested genotypes were: M-25, B-10-50, MAK-1-79, M-25-54, MAK-1, M-25-58, B-10 and MAK-1-60. Higher accumulation of ³⁵S was recorded in B-10-50, MAK-1 and M-25-58 than the other genotype. The accumulation of 35S in the seed was higher in M-25-54 and MAK-1. This indicates that these genotypes have high ability to transport of S from root to shoot and seed compared to other genotypes. Moreover, transport rate of 15N and the accumulation of total nitrogen was also higher in these genotypes of B-10-50, MAK-1 and M-25-54.

Objectives:

To find out physiological causes of poor yield in blackgram for with and without S and N.

Methodology:

Sand culture experiments were conducted at the pot yard of Bangladesh Institute of Nuclear Agriculture, Mymensingh. Eight blackgram genotypes were evaluated on the basis of S and N uptake capacity. The eight genotypes were: M-25, B-10-50, MAK-1-79, M-25-54, MAK-1, M-25-58, B-10 and MAK-1-60. The crops were grown in acid washed sand in the pots lined with polythene bags and in the plastic pots in hydroponic condition. For sand culture experiment, sterilized seeds were sown in the pots. Seven-day after germination extra seedlings were uprooted and 4 plants were kept in each pot. Two sets of pots for each crop were maintained. Of them, plants of one set were kept undisturbed (1^st set) for yield purpose while the other set (2^nd) was used for analytical purpose. Plant from the second set biochemical analysis such as nitrate reductase activity, chlorophyll, soluble protein, reducing sugar were done. Four rates of sulphur viz., 0, 10, 20 and 40 kg S/ha were used. Nitrogen @ 30 kg/ha was used to each of the S dose. In these pots, 15N and 35S were applied at pod filling stage. To avoid S-effects on plant growth, S was not applied in the control pots. Seeds of these plants were analyzed for 15N and 35S. For hydroponic culture, pre-germinated seeds were placed in pots containing 0.1 strength nutrient solution and maintained up to two weeks. The ¹⁵N and ³⁵S were applied in these pots for their uptake study. Both ¹⁵N and ³⁵S were analyzed 48 hours after application. All the data were subjected to statistical analysis.

Achievement/Findings:

The genotypes which have greater metabolic activities and transport of ¹⁵N and ³⁵S also have high yield potentiality.
