Screening of Summer Mungbean Genotypes Based on Sulphur and Nitrogen Uptake and Transport

Abstract/ Executive Summary:

Pot culture experiments were conducted to screen high nutrients uptake genotypes of summer mungbean. The tested genotypes were: E₄I-915, N₅I-521, Kanti, E₁I-901 and Binamung-2. S level increased the accumulation of N and S. Variability was recorded in the tested genotypes with respect to sulphur and nitrogen uptake. Binamung-2 had the lowest accumulation of ³⁵S while that was the highest in advanced line E₄I-915. The genotypes E₄I-915 and N₅I-521 accumulated comparatively higher sulphur both in shoot and seed indicating better translocation efficiency of sulphur in these two genotypes. Moreover, transport rate of ¹⁵N and the accumulation of total nitrogen were also higher in the genotypes of E₄I-915 and N₅I-521.

Objectives:

To find out physiological causes of poor yield in summer mungbean for unavailability S and N.

Methodology:

Sand culture experiments were conducted at the pot yard of Bangladesh Institute of Nuclear Agriculture, Mymensingh. Five summer mungbean genotypes were evaluated on the basis of S and N uptake capacity. The five genotypes were: E₄I-915, N₅I-521, Kanti, E₁I-901 and Binamung-2. The seeds were grown in acid washed sand in the pots lined with polythene bags and in the plastic pots in hydroponic condition. For sand culture experiment, sterilized seeds were sown in the pots. Seven-day after germination extra seedlings were uprooted and 4 plants were kept in each pot. Two sets of pots for each crop were maintained. Of them, plants of one set were kept undisturbed (1^st set) for yield purpose while the other set (2^nd) was used for analytical purpose. Plant from the second set biochemical analysis such as nitrate reductase activity, chlorophyll, soluble protein, reducing sugar were done. Four rates of sulphur viz., 0, 10, 20 and 40 kg S/ha were used. Nitrogen @ 30 kg/ha was used to each of the S dose. In these pots, ¹⁵N and ³⁵S were applied at pod filling stage. To avoid S-effects on plant growth, S was not applied in the control pots. Seeds of these plants were analyzed for ¹⁵N and ³⁵S. For hydroponic culture, pre-germinated seeds were placed in pots containing 0.1 strength nutrient solution and maintained up to two weeks. The ¹⁵N and ³⁵S were applied in these pots for their uptake study. Both ¹⁵N and ³⁵S were anlyzed 48 hours after application. All the data were subjected to statistical analysis.

Achievement/Findings:

The genotypes which have greater metabolic activities and transport of ¹⁵N and ³⁵S also have high yield potentiality in summer mungbean. The genotypes E₄I-915 and N₅I-521 accumulated higher rate of S and N, and showed higher seed yield.
