Effect of Bio Agents on Growth and Root Rot of Lentil

Abstract/ Executive Summary:

Laboratory and pot experiment was conducted to control root rot caused by Fusarium solani and Sclerotium rolfsii, a devastating and wide spread disease in the world. Ten bio-agents including control were tested in these experiments. Macerated extract of Fusarium solani + Trichoderma harzianum showed the best result in controlling root rot of lentil with the highest seed germination (100%), radical length (1.56 cm), seedling emergence (95.73%), root length (8.16 cm), shoot length (17.75 cm), number of branches/5 plants (15.56). The lowest root rot (25.93%) was also observed in the treatment of Fusarium solani + Trichodrma harzianum. The other four treatments, macerated extract of Sclerotium rolfsii, Fusarium solani, Trichoderma harzianum, Sclerotium rolfssi + Trichoderma harzianum showed better results over control in terms of all plant parameters. Leaves of Jarul, Mehegoni and Acacia showed poor performance compared to control in almost all parameters.

Objectives:

To evaluate different bio agents in creating induced resistance against root rot pathogen and enhance growth of lentil

Methodology:

The experiments were conducted at the Laboratory and pot yard of Bangladesh Institute of Nuclear Agriculture during November 2011 to March 2012 using Binamasur-2, the susceptible mutant variety to collar rot. This study consisted of ten treatments macerated extract of Sclerotium rolfsii, Fusarium solani, Trichoderma harzianum, Sclerotium rolfsii + Trichoderma harzianum, Fusarium solani + Trichoderma harzianum, extract of Jarul (Lagerstroemis sp.) leaf, Mehegoni (Swietenia sp.) leaf, Acacia (Acacia sp.) leaf and chemical fungicide, Provax (0.3%) [Carboxin 37.5% + Thirum 37.5%] and untreated control. To prepare interaction culture of Sclerotium rolfsii and Trichoderma harzianum, a large petridish was taken and earlier prepared the pure culture of the two organisms were placed into two sides of the petridish. All these operations were done in the laminar air flow chamber. About 10 plates were prepared and incubated for growth. When the two organisms came close contact of each other then it was collected by scraping and macerated extract was prepared. The same procedure was followed to prepare interaction culture of Fusarium solani and Trichoderma harzianum. Disease incidence were taken after 10, 20, 30 and 40 days after sowing.
