Induction of Stem Rot Resistant Mutants of Jute Through Nuclear Techniques

Abstract/ Executive Summary:

The resistance against Stem rot disease of jute induced in Corehorus capsularis and C. olitorius mutants through gamma-irradiation of seeds. Among the induced mutants, CM-36 developed from the cultivar D-154 was found to be resistant during early generation and becomes susceptible at the advanced generations. The mutant cultivar Atompat-38 was found to be moderately resistant to stem rot. The mutant lines OM-70, OM-42, OM-40 and the cultivars O-4 and O-9897 were moderately resistant to stem rot during early stage of plant growth but become susceptible at plant maturity. Atompat-38, OM-51, CG, CVL-1, OM-37, OM-10 and OM-1 were moderately resistant in both early and advanced stages of crop growth.

Objectives:

To identify the induced mutants of jute against Stem rot.

Methodology:

Experiment 1: Dry seeds of the cultivar D-154 were exposed to different doses (70, 80, 90 and 100 KR) of gamma-irradiation from ⁶⁰Co sources. The induced mutants identified during M₂ and subsequent advanced generations were screened against Macrophomina phaseolina under artificially inoculated conditions in the glasshouse following Mia et. al. 1974 and 1976: Shaikh et al. 1977a and 1977b and Mia and Shaikh, 1966.

Experiment 2: Pre-soaked seeds of the jute vultivar CVL-1 were treated with 3, 6, 9, 10, 12, 15, 20, and 25 mm concentrations of the chemical mutagen N_aN₃ in 1984 (Anonymous, 1984). The induced mutants selected from M₂and the subsequent generations along with a hybrid line C-443 (Atompat-38 x CVL-1) were screened against the isolate (s) of Macrophomina phaseolina during 1995 and 1996 jute growing seasons. The induced mutants selected from experiment 1 and 2 were evaluated under field condition in 1997 jute growing season at the Bangladesh Institute of Nuclear Agriculture Farm, Mymensingh. The experiment was conducted in a split-plot design with three replications. The isolates Macrophomina phaseolina were tested in the main plots and the induced mutants/cultivars of jute were tested in the sub-plots.
