Analysis of Genetic Relationship Among Indigenous Sheep Population in Bangladesh

Abstract/ Executive Summary:

Sheep is a very important and promising animal genetic resource of Bangladesh. No recognized sheep breed is available. for this a research has been undertaken with the objective to evaluate the genetic relationship among indigenous sheep populations in Bangladesh, to compile available research data and to select exotic breed (s) suitable for crossing with indigenous sheep of Bangladesh from available research findings of different findings. Around 96 blood samples were collected from Barind, Jamuna River Basin, Coastal, sheep of Sylhet division, Garole and Chotanagpuri sheep of India from Meherpur district of Bangladesh. The Garole and Chotanagpuri breeds were used as reference breeds for this study. DNA was extracted quantified from blood samples using commercially available DNA extraction kit as per manufactures' protocol. DNA was quantified using a nanodrop machine. DNA processing a 280/260 ratio of 1.7 to 1.8 was used for PCR analysis. Then information were collected through reviewing from different journals, books, proceedings, annual report, post graduate thesis and internet search. Genetic distance between Jamuna river basin and Barind was lowest (0.0891) and between Garole and Costal was highest (0.1786).

Methodology:

Around 96 blood samples were collected from Barind, Jamuna River Basin, Coastal, sheep of Sylhet division, Garole and Chotanagpuri sheep of India from Meherpur district of Bangladesh. The Garole and Chotanagpuri breeds were used as reference breeds for this study. DNA was extracted quantified from blood samples using commercially available DNA extraction kit as per manufactures' protocol. DNA was quantified using a nanodrop machine. DNA processing a 280/260 ratio of 1.7 to 1.8 was used for PCR analysis. The PCR were conducted with 16 microsattelite markers and the PCR products were sent to China for fragment length analysis. Then information were collected through reviewing from different journals, books, proceedings, annual report, post graduate thesis and internet search. Total 53 papers/reports/proceedings on indigenous sheep of Bangladesh were collected for this studies. This review document covered current sheep population and their trend in Bangladesh, historical perspective and their uses, production and reproduction performances, genetic characteristics, health problems and other important information.

This study was conducted to evaluate the genetic relationship among indigenous sheep populations of Bangladesh (Barind, Jamuna river basin, Coastal and Garole sheep) using microsatellite markers. A total of 96 blood samples were collected from adult sheep of (i) Barind (24), (ii) Jamuna River Basin (24), (iii) Coastal (24), iv) Garole (10) and (vi) Chotanagpuri (10) sheep of India. Chotanagpuri sheep available in the Meherpur district of Bangladesh was used as a reference breed. DNA was extracted and quantified from blood samples. Genetic distance between Jamuna river basin and Barind was lowest (0.0891) and between Garole and Costal was highest (0.1786). Garole and Chotonagpuri sheep has higher genetic distance from other three sheep populations. Phylogenetic dendogram showed that sheep of Jamuna river basin and barind belong to same genetic group. Whereas, coastal, garole and Nagpur sheeps were shown higher genetic distances from Jamuna river basin and coastal sheep. Considering findings of this study it may be concluded that the Barind and Jamuna river basin sheep belongs to a similar genetic group while, Garole and coastal sheep are belonging to two distinct genetic groups.
