Studies on the Quality of BINA Tomato-5 Seeds Kept in Different Containers

Abstract/ Executive Summary:

Seed samples of Binatomato-5 were studied for germination, seedling vigour and health after 25, 50 and 75 days of storage in three different containers, namely metal container, polythene ban and small brown paper bag. Among the three containers, the best performance was observed when the seeds were stored in polythene. Fusarium oxysporum, Aspergillus niger, Aspergillus flavus and Penicillium spp. were found to be associated with samples. The prevalence of these pre-dominant fungi were found lower in polythene bag.

Objectives:

To assess the effect of different containers on the seed quality of Binatomato-5.

Methodology:

The storage and laboratory experiments were conducted during April 2006 to June, 2006 at Seed Pathology Centre, BAU, Mymensingh. The seed samples of Binatomato-5 and three different types of containers viz. small brown paper bag, polythene bag and metal container (small powder milk container) were used. The collected seed samples were stored in sore room of the Seed Pathology Centre, BAU, Mymensingh, Bangladesh. Tests were conducted at 25, 50 and 75 days after storage (DAS).Germination test of the stored sample was done in sand using tray method. Plastic trays (18" x 9") were used for the purpose. 100 seeds were sown in sand in each plastic tray in 4 lines. For each sample altogether 400 seeds were tested for germination. Germination was recorded at 14 days after sowing. Normal seedlings, abnormal seedlings and ungerminated seeds in terms of fresh seed, hard seed and death seed were counted separately and expressed in percentages. While evaluating the number of normal seedlings at the time of final count, the seedlings length of 5 randomly selected seedlings were also measured. Vigour index was calculated by multiplying germination (%) with seedling length (mm) of the same seed lot. The seed lot showing higher vigour index was considered to be more vigorous (Gupta, 1993). Seed health test was carried out following blotter method (ISTA, 1996).
