In Vitro Regeneration and Bulblet Formation in Garlic

Abstract/ Executive Summary:

Present study was conducted to establish an efficient protocol for plantlet and bulblet formation through embryogenesis in garlic. Root tips were excised from aseptically sprouted garlic and cultured on MS medium containing different concentrations and combinations of 2,4-D and BAP for embryogenic callus formation. The most favourable concentration was I mg/l 2,4-D supplemented with 5 mg/l BAP for callus and embryo formation. Embryos germination and highest number of bulblets formation were observed in MS medium supplemented with 10 mg/l BAP. Rooting of the individual plantlet with bulblet was induced after transfer to medium without growth regulator. In vitro bulblets were obtained from root tip via embryogenic callus. The plantlets were established in the soil after acclimatization.

Objectives:

To develop suitable protocol for in vitro regeneration of garlic via embryogenic callus and in vitro bulblet formation to produce rapidly true to type propagules.

Methodology:

The experiment was conducted in the tissue culture laboratory, department of Genetics and Plant Breeding, Bangladesh Agricultural University, Mymensingh in a CRD two factors experiment with five replications. Three garlic genotypes including one exotic and two local namely "Burma", ''Faridpur" and "Haluaghat" were used in this trial. Healthy cloves were surface sterilized with seventy percent alcohol for thirty seconds and then 0.1 % H_gC_I2 solution with 2 drops Tween-20 per 100 ml for minutes. Subsequently those were rinsed three times by sterilized distilled water. Root tips of sprouted cloves about 2-3 mm long were used as explants. Explants were placed on MS medium supplemented with different concentrations (0, 1.0, 1.5, 2.0 mg/l) of 2,4-D with the combination of BAP (0, 5.0, 10.0, 15.0 mg/l) for callusing. Fourty five days older calluses were sub-cultured in regeneration and bulblet formation media. For this purpose MS medium in combination with different concentration (0, 5, 10 mg/l) of BAP were used. Data on different parameters were collected and statistical analysis was performed by MSTAT programme following CRD.
