POST GRADUATE CERTIFICATECERTIFICATE COURSE ON SEED TECHNOLOGY
Department of Agronomy, Sher-e-Bangla Agricultural University, Dhaka- 1207

The experiment was conducted at Sher-e-Bangla Agricultural University during January to July 2006 to determination of the effect of storage container on seed quality and to observe deterioration of germination capacity of seed in respect of time due to biotic and abiotic factor in different storage container. The initial moisture content of seeds in tin container, gunny bag and polythene bag were similar (10.6, 10.7 and 10.5%) but it was increased with increasing storage time. The initial germination percentage of seeds in tin container, gunny bag and polythene bag seeds was 94.0, 87.67 and 88.67 percent but after storage it was declined. The decline rate is higher in seeds of gunny bag. Very strong negative relationship was observed between germination percentage and moisture content of chilli seeds stored in every container.

1. To determination of the effect of storage container on seed quality.

2. To observe deterioration of germination capacity of seed in respect of time due to biotic and abiotic factor in different storage container.

The experimen was conducted at Sher-e-Bangla Agricultural University during January to July 2006 to determination of the effect of storage container on seed quality and to observe deterioration of germination capacity of seed in respect of time due to biotic and abiotic factor in different storage container. Chilli seeds were collected from Agronomy department of Sher-e-Bangla Agricultural University. Seeds were kept in three types of container such as tin pot, poly bag and gunny bag and stored at room temperature and RH for around 2 months. Collected seeds were compared for initial moisture content, seed size, germination and seedling vigor. Moisture content was determined by using oven dry method in the Agronomy laboratory of SAU. Seeds from each 3 containers were taken and moisture content was measured using moisture meter. For germination test one hundred seeds of each sample were placed in petridish containing blotting paper soaked with water. For each test, three petridishes were used. The petridishes were placed in room temperature for 7 and 14 days for germination. Seedlings were counted every day up to the completion of germination at 7 and 14 days. A seed was considered to be geminated as seed coat ruptured,plumule and radicle came out up to 5 mm in length. Germination percentage was calculated using the following formula (Krishnasamy and Seshu, 1990).Germination (%) =  Number of seed germinated/Number of  seeds Tested  X 100 Seeds were stored at room condition in different containers and germination test was performed every 14 days. Seedlings obtained from standard germination test were used for seedling evaluation. Normal or abnormal seedlings were classified according to the rules of ISTA (1993). Germination rate (GR) was calculated from a daily count of germinated seeds until it reached a constant value, using the following formula given by Maguire (1962).

Number of normal seedlings

GR=   Number of normal seedlings/ Days to first count    +………..….+ Number of normal seedlings/ Days to Final count

Germination index (GI) was calculated using the following formula given by Scott et al. (1984).

GI= ΣTiNi/S

Where,

Ti= number of days after sowing

Ni= number of seeds germinated on day i

S= total number of seeds placed in the petridish

The coefficient of velocity (CV) was calculated using the following formula given also by Scott et al. (1984).

CV = ΣNi/ΣNiTi x 100

Where,

Ti= number of days after sowing

Ni= number of seeds germinated on day i

For recording weather data, Reading of dry bulb and weight bulb temperature and relative humidity of inside the room was recorded at 8 am and 6 pm in every day begging from collection to 50 days after storage.For observation of biotic factor that influence seed quality, 250 g of seeds were taken and observed the numbers and kind of insects or pathogen were presented.
