Plant Regeneration from Anther Cultures of Some F1 Hybrids of Rice

Abstract/ Executive Summary:

Anthers of F₁s of seven cross combinations between salt tolerant lines and high yielding rice varieties were used in an attempt to induce callus and regeneration of green plants, Chaleff s R2 medium supplemented with 2.0 mg/1 NAA + 0.5 mg/1 kinetin was used for callus induction F₁ hybrids of all the cross combinations produced calli with frequency ranging from 1.78 to 7.71%. The highest frequency of callus formation was found in Binnatoa x BR9 combination. Plantlet regeneration occurred when calli were transferred to MS medium supplemented with 1.0 mg/1 1AA +1.0 mg/1 kinetin. Calli of Binnatoa x BR9, Pokkali x 1R5657 x BR 11 and 1R21015 x BR11 produced both greens and albino plants. Maximum yield of green plantlets was observed in Binnatoa x BR9.

Objectives:

To create of F₁ hybrids of seven cross combinations of rice were studied for their regeneration ability via callus formation.

Methodology:

In the present study three HYVs e.g BR9, BR11, BR14 and four salt tolerant varieties e.g Binnatoa, Pokkali, N. Bokra, IR5657 and IR21015 were used. During 1992, F₁ seeds of seven combinations namely, Binnatoa x BR9. Pokkali x BR11. Pokkali x IR21015. Pokkali xBR14, N. Bokra x BR11, IR5657 x BR11, and IR21015 x BR11 were developed. The F₁ seeds after surface sterilization with 70% ethyl alcohol were germinated in Petri dishes. Seedlings were transferred in earthen pots. Three-week-old seedlings were transplanted in the field using one seedling per hill. F₁ plants were selected and boots with an auricle distance of 5-10 cm between the flag-and the nest leaf were collected between 8 and 10 a.m. Boots were washed with tap water, swabbed with 70% ethyl alcohol and ten subjected to cold pretreatment for 7-10 days at 10⁰C ( Chen et al. 1991). After cold treatment boots were surface sterilized by means of 70% ethyl alcohol. Anthers were collected from mid portion of the panicles. They could be easily removed by cutting spike lets at their base and then tapping them directly in the wide- mouthed 100 ml conical flasks. Anthers derived from panicles as mentioned above had a very high population of mid-uninucleate pollen. Anthers were plated on Chaleff’s R2 medium (Chaleff 1980) supplemented with 2.0 mg/1 NAA ± 0.5 mg/1 kinetin and incubated in dark at 25 ± 1⁰C till callus induction. Calli produced in 40, 50 and 60 days of incubation were transferred to MS (Murashige and Skoog 1962) medium supplemented with 1.0 mg/1 IAA + 1.0 mg/1 kinetin + 50 g/1 sucrose and incubated at 26 + 1⁰C under 16 hr photoperiod (2000 lux). As regeneration did not always result in complete plants i.e. shoots and roots together, shoots without well developed roots were transferred to half-strength MS + 0.5 mg/ 1 NAA + 2.5 mg/1 MET (Sun and Zhao 1992) and kept there till well developed roots were evident. All the regenerated plants were transferred to earthen pots.

Achievement/Findings:

Calli of Binnatoa x BR9, Pokkali x IR21015, IR5657 x BR11 and IR21015 x BR11 produced both green and albino plants. Maximum yield of green plantlets were observed in Binnatoa x BR9.
