Chromosome Banding in Two Species of Corchorus and Their F1 Hybrid

Abstract/ Executive Summary:

Number of bands and banding patterns from prophase chromosomes of two species of Corchorus and their F₁ hybrid was determined by HCI-Giemsa technique. The bands were produced clearly with 4% Giemsa diluted with Sorenson buffer solution. Both terminal and interstitial bands were observed. Unlike C. capsularis all the pairs of chromosomes in C. olitorius were homomorphic. Banding patterns of three chromosomes in F₁ were found to be common to those of C. capsularis and C. olitorius.

Objectives:

To study banding patterns in two species of Corchorus and their F₁ hybrid by HCI-Giemsa technique.

Methodology:

Two species of Corchorus such as C. olitorius L. var. 0.4, C. capsularis L. var. D-154 and their F₁ hybrid were obtained from the Plant Breeding and Tissue Culture Lab, Department of Botany, University of Dhaka. To study banding pattern in two Corchorus species and their F₁ hybrid, actively dividing root tips were pretreated with saturated solution of paradichlorobenzene (PDB) for 2-4 hours at 10⁰C, fixed in aceto- alcohol (1:3) for 48 hours and stored in 70% alcohol. HCI- Giemsa technique was performed according to the schedule of Joshi and Ranjckar (1980) to band the chromosomes. Prior to treatment with 8N HCI for 10 minutes slides were incubated in water for 10-15 minutes. Slides were then rinsed thoroughly with cold distilled water to stop hydrolysis abruptly, and were stained in 4% Giemsa (BDH) diluted with M/15 Sorenson’s phosphate buffer at pH 6.8 After air drying the preparations were mounted in cuparol mounting medium.
