Regeneration of Six Oryza Sativa L. Indica Salt Tolerant Varieties from Mature Embryos.

Abstract/ Executive Summary:

Regenenration capacity of callus cultures induced from mature seeds of six indica varieties was tested. The indica varieties were chosen due to their ability to grow in saline soils of regions of Noakhali ( ECe 3-12 dS/m). The Aus or dry season varieties chosen were Chiknal. Binnatoa and Boilam and the Transplanted Aman varieties were Rajashail,, Nona Balam and Patnai-23. By including BAP, Yeast Extract and decreasing the amount of 2. 4-D in the MS induction medium, it was possible to increase regeneration up to 37 -41% in Nona Balam, 30-35% in Rajashail and 11-13% in Patnai-23. Addition of tyrosine and IAA in the medium after induction of calli of the Aus varieties, resulted in 30-33% regeneration in Binnatoa, 24-28% in Boilam and 22-25% in Chiknal.

Objectives:

To crate varieties with good regeneration capacity. Once identified suitable varieties can be chosen for the production of salt tolerant lines through anther culture

Methodology:

Six indica rice varieties which farmers grow in saline areas of Noakhali and Barisal namely, the Aus varieties Binnatoa, Boilam and Chiknal and the T. Aman varieties Nona Balam, Rajashail and Patnai-23 were used. All seeds were kindly provided by Bangladesh Rice Research Institute (BRRI). Fully mature and healthy seeds were dehusked and surface sterilized in 3% sodium hypochlorite solution for 15 minutes and rinsed 3-4 times with sterile distilled water. To induce callus in the mature embryos MS (Murashige and Skoog 1962)media supplemented with 2, 4-D (2 mg/1). Yeast Extract (YE) (3 g/1), sucrose (30 g/1) and 0.7% agar was used according to Miah et al. (1987). After 21 days in the dark at 24 + 10⁰C, the callus was transferred to MS subculture medium containing 0.5- 1 mg/ 12, 4-D, 1mg/ 1 BAP according to Brar et al. (1988) with modifications. Callus was kept in light for 12 hour cycles. After 21 days in subculture medium, 0.5 cm pieces of callus were transferred to MS regeneration medium containing 0.37 mg/ 1 NAA, 10.7 mg/1 kinetin, 3 g/ 1 YE, 3 g/1 Casein Hydrolysate (CH),70 g/1 sucrose, under light according to Miah et al. (1987). Modifications had to be made to the regeneration media to achieve regeneration in some varieties (See results section). Regenerated plants were transplanted from test tubes to sterilised soil after 3-4 weeks and covered with polythene bage to prevent wilting. Results shown are an average of 3 replications. For each replication 50 seeds of each variety were inoculated. Each subculture tube generated enough calli for transfer to 2-3 test tubes containing the regeneration medium.
