Prevalence of Enteric Bacteria Isolated from Cloacal Swabs in Sonali Chickens

Abstract/ Executive Summary:

An attempt was undertaken to determine the prevalence of enteric bacteria in Sonali chickens (Fayoumi hen x RIR cock). For this, 100 cloacal swabs were collected from apparently healthy chickens in sterile nutrient and tetrathionate broth. Isolation and identification of the bacteria were performed by culturing in different media, staining and biochemical tests. Five different bacteria viz. Salmonella, E. coli, Staphylococcus, Streptococcus and Pasteurella multocida were isolated from cloacal swabs and their prevalence were 29% (29/100), 91% (91/100), 64% (64/100), 46% (46/100) and 2% (2/100), respectively. The lower prevalence of fowl cholera was assumed to be due to the use of fowl cholera vaccine.

Objectives:

To determine the prevalence of bacterial flora in the cloaca of Sonali birds.

Methodology:

100 Cloacal swabs were collected from apparently healthy Sonali chickens in sterile test tube containing sterile nutrient and tetrathionate broth uing sterilized cotton tipped swab sticks from SLDP-2 area, Feni. The tubes were placed in icebox and transferred to the bacteriological laboratory, Department of Pathology, Bangladesh Agricultural University, Mymensingh. The broth containing swabs in the test tube were incubated at 37°C for 24 hours for the growth of bacteria. Then they were plated on different culture media and subcultured following standard procedure for the isolation of specific bacterial colonies. The isolated bacteria were stained with modified Gram's and Leishman's stains as described for study their morphology. The isolated bacteria were subject to biochemical tests using different sugars and chemicals. Methyl Red, Voges-Proskauer, Indole, Catalase and Coagulase were performed for isolation and identification of the organisms following the procedures described. Young broth culture of the organisms, incubated at 37°C or below the optimum temperature (22°C), was examined in 'hanging drop' preparation, using a high power oil immersion objective with reduced illumination.

Achievement/Findings:

The present study showed that though the organisms were present in the cloaca of apparently healthy Sonali chickens, they did not produce clinical diseases. Since it seems that some other factors are involved with the clinical disease production yet to be uncovered.
