Effect of Brine Solution on the Seed Borne Disease of Rice

Abstract/ Executive Summary:

An investigation was carried out to know the efficacy of brine solution on the control of seed borne pathogen. The experiment was conducted at plant pathology laboratory, BRRI, Gazipur throughout season-2014. The experiment was conducted following Completely Randomized Design (CRD) with three replications. Six treatments in different concentration like B₁₀₀, B₅₀, B_30,B_20,B₁₀ and B₀control of 2% brine solution (200gm salt in 1 liter of water) were used to see the effect of brine solution against seed borne disease of rice such as Bipolaris, Aspergillus, Sarocladium, Penicillium, Pyricularium, Rhyzopus, Fusrium etc. Susceptible variety BRRI dhan33 as basic seed collected from Petrochem Seed Company were seeded in different Petri dishes with different concentration by maintaining three replications. The seeds were dipped in different concentration of brine solution for 24 hours, after that, the seeds were kept to dry for 15 minutes and were plated on PDA for 1 week to germinate. The petridishes containing seeds were incubated at 20±2° under alternating cycles of 12h near ultraviolate (NUV) and darkness for 7 days. After 7 days of incubation the seeds were observed for the presence of seed-borne fungi Bipolaris, Fusarium etc. under stereo binocular microscope. Germination% of the seeds also was recorded. The highest germination was observed in 10% brine solution (B₁₀). However, brine solution has no significant effect on seed germination. Shoot length increased up to 30% brine solution but no consistent result was found in root length. The incidence of Fusarium, Bipolaris and Curvularia were detected as 2-7, 0-14 and 9-26% in treated seeds. Other pathogens were detected very less amount (<3%) in both treated and untreated seeds. The percent infection of Bipolaris decreased significantly except for 30% brine solution. Incidence level of Fusarium also decreased but no consistency of the incidence was observed among different treatments. Further, different treatments had no effect on Curvularia. Further investigation will be needed to conclude the results precisely.
