Study on the Pathogenecity Level of Fusarium Solani and Sclerotium Rolfsii Causing Root Rot and Collar Rot of Lentil

Abstract/ Executive Summary:

Two experiments were conducted to evaluate the level of pathogenecity of Fusarium solani causing root rot and Sclerotium rolfsii causing collar rot of lentil (Lens culinaris Medik). F. solani and S. rolfsii were collected from infected pulse crops of lentil, chickpea, blackgram, groundnut and mungbean at nine different disease prone locations of Bangladesh. Growth and phenotypic character of F. solani and S. rolfsii were recorded. Linear mycelial growth of the different isolates of F. solani were ranged from 77.9 to 89.8 mm and shape, colony and texture were more or less regular, whitish-cream and floccose, respectively. The highest linear mycelial growth of S. rolfsii was found 90.02 mm in the isolate of SL-4. Most of the isolates were radiate in growth pattern, faster growth habit and brownish in colour. The pathogenecity level of F. solani and S. rolfsii were studied under pot culture condition. The lowest germination (70%) and the highest pre-emergence death (30%) of lentil were found in the isolate FL-4. The highest root rot (53%) and the lowest plant stand were found in the isolate FBg-1. In case of S. rolfsii, the lowest germination (29.8%) and the highest pre-emergence death (70.3%) were found in the isolate SL-4. The highest collar rot (16.0%) and the lowest plant stand (13.8%) were found in the same isolate. The isolates FL-4 and FBg-1 of F. solani and the isolate SL-4 of S. rolfsii were used to evaluate the mutants/strains of lentil against the major soil borne diseases of root rot and collar rot.

Objectives:

To collect, isolat, purification, identification and preservation of isolates of Fusarium solani and Sclerotium rolfsii

Methodology:

The laboratory work and pot experiments were carried out in the laboratory of Plant Pathology Division and shade house of Bangladesh Institute of Nuclear Agriculture (BINA), Mymensingh, Bangladesh during the period of 2010. Diseased plant samples of root rot and collar rot were collected from different cropping area of Bangladesh. Then isolation, identification, purification, preservation and growth behavior of Fusarium solani and Sclerotium rolfsii were done at laboratory. The pathogenecity was tested at pot. Fusarium solani and Sclerotium rolfsii were isolated from different diseased plant samples collected from different locations of Bangladesh. The samples showing typical root rot symptoms were brought to the laboratory of Plant Pathology Division, BINA, Mymensingh. The samples were washed initially with tap water to remove sand and soil particles and cut into small pieces (1.0 cm) along with healthy and dead tissues. Surface sterilization was done with 1:1000 mercuric chloride solution for 1 minute. Then the plant pieces were washed thrice with sterilized water and placed on filter paper to remove excess water adhering to the pieces. Thereafter, five pieces were plated in PDA plates aseptically maintaining equal distances (Begum et al. 1998). The plates were incubated at 28 ± 1 ⁰C temperature for 7 days and observation was taken regularly to see the growth of fungi from the plant pieces. The fungus was then purified by hyphal tip culture technique (Tuite 1969). The isolated fungus was identified following the key outlined by Booth (1971) and Singh (1982). The pure culture of the isolates of Fusarium solani and Sclerotium rolfsii were preserved in PDA slants at 5 ± 1 ⁰C in refrigerator as stock culture for future use (Begum, 1997).
