Identification of Male Sterility Maintainer Lines for Ogura Radish (raphanus Sativus L.)

Abstract/ Executive Summary:

Ogura radish', a cytoplasmic genetic male sterile line, was crossed with four local and three Japanese cultivars to identify maintainer lines. Out of seven F₁ families, one cross involving a local cultivar, Aushi, produced 100% male sterile (MS) progeny. The crosses involving the other two local cultivars, Tangail Local and Kuni, produced about 90% MS progeny, indicating the presence of maintainer gene(s) for male sterility. The fourth local cultivar, Tasaki, produced 100% male fertile (MF) progeny. All three exotic cultivars appeared to possess the chromosomal gene(s) for controlling the male sterility. In BC₁ BC₂ and BC₃ generations, segregation of MS plants were more frequent when 'Aushi' was used as recurrent parent. The expression of male sterility was not affected by seasonal influences. Thus the local cultivar 'Aushi' may be used as maintainer line for 'Ogura radish'. To produce hybrid seed, 'Tasaki' can be used as pollinator line as it exhibit high heterosis with 'Aushi'.

Objectives:

To identify maintainer lines with local adaptation for 'Ogura radish' with a view to develop hybrid varieties.

Methodology:

Seeds of 'Ogura radish' and its maintainer line 'Normal' were collected from Professor Ogura of Kagoshima University, Japan in July 1991. Sprouted seeds of 'Ogura radish', 'Normal' and two other Japanese radish cultivars (Japanese White and Osaka) were vernalized at 5°C for four weeks as these cultivars do not flower or poorly flower under Bangladesh conditions. The four male fertile inbred lines of local radish viz. Tangail Local, Aushi, Kuni and Tasaki and the three exotic inbred lines viz. Japanese White, Osaka and Normal were grown at the same time. At flowering the 'Ogura radish' was crossed with these lines during the winter season of 1991-92 at the Institute of Postgraduate Studies in Agriculture (IPSA), Salna, Gazipur 1703, Bangladesh. Seven F_I families were grown in both pot and in the field during the winter season of 1992-93. The plants of all the crosses flowered without vernalization. The progenies of the crosses involving the local cultivars flowered earlier compared to their mother parent. Performance of MS plants of FI families in respect of plant height (cm), days to bolting, days to flowering, days to pod maturity and seed yield per plant (g) were recorded from ten randomly selected plants. In addition few F_IMS plants of the cross 'Ogura radish' x 'Tangail Local' and 'Ogura radish' x 'Aushi' were backcrossed with their parents.

First backcross (BC_I) populations of the two crosses were grown during summer, 1993. Few MS plants of BC₁ families were backcrossed with the MF parents to get BC₂ families. Second backcross (BC₂) and third backcross (BC₃) generations of the two crosses were evaluated during winter, 1993-94 and summer, 1994. At flowering both MS and MF plants were identified by staining the pollen grains with 1% acetocarmine at the three different stages of flowering such as beginning of flowering, the full bloom and the late flowering. Plants possessing all flowers with shrivelled anthers, which did not produce viable pollen, were scored as MS.
