Salt Tolerance of Three Barley Cultivars at Early Growth Stages

Abstract/ Executive Summary:

Three cultivars of Barley were tested at the physiology Laboratory of BARI, Joydebpur in the year 1992 for salt tolerance at germination and early seedling stage. Four salinity gradients, 4.0, 8.0, 12.0 and 16.0 dS^-m sodium chloride solution were used against distilled water (control for germination) and with Hoagland nutrient solution for seedling stage. Germination, shoot and root lengths, dry weights of shoot and root were significantly decreased at 16.0 dS^-m. Although gerrnination was stimulated for all the cultivars at 8.0 dS^-m, BARl Barley-1 showed a decrease in shoot and root dry weight which have also been reflected in Na⁺, K⁺ uptake and accumulation in Barley plants.

Objectives:

To find out the relative salt tolerance of three cultivars including BARl Barley-1 up to seedling stage.

Methodology:

The seeds of three cultivars of barley viz. BARl Barley-1, Centinella and API-19 were used for this study. The germination was carried out in petridishes according to ISTA (1985) rules. Five levels of salinity, distilled water (control), 4.0, 8.0, 12.0 and 16.0 dS^-m of sodium chloride solution were used for germination test. For seedling growth, 4 inches diameter plastic pots were used. Treatments were as follows: C₁= distilled water mixed with Hoagland nutrient solution (0.4 dS^-m) at 25°C. C₂, C₃, C₄ and C₅= salt of sodium chloride added to distilled water and Hoagland nutrient solution to prepare 4.0, 8.0, 12.0 and 16.0 dS^-m salinity levels. Twenty seeds were placed on the top of the perforated cover of the pot filled with treatment solution and replicated thrice. The solution was changed first at four days and then at 3 days interval. The temperature was maintained at 22°C±2°C. Length (cm) and dry weight (g) of root and shoot of 10 seedlings per replicate were recorded after 14 days. Ions (Na⁺, K⁺and CI‾) were extracted by boiling the tissue with distilled water. Na⁺ and K⁺ions were measured using a Flame Analyzer and CI‾ ion was measured by standard titrametric method using potassium chromate as an indicator. Analysis of tissue for ion accumulation were done for control, 8.0 and 16 dS^-m treatments.

Achievement/Findings:

Germination, shoot and root lengths, dry weights of shoot and root were significantly decreased at 16.0 dS^-m. Although gerrnination was stimulated for all the cultivars at 8.0 dS^-m, BARl Barley-1 showed a decrease in shoot and root dry weight which have also been reflected in Na⁺, K⁺ uptake and accumulation in Barley plants. Our experiments therefore indicate that BARI Barley-1 is the most sensitive among the three barley cultivars.
