Effects of Oral Administration of Glucocorticoids on Some Haemato-biochemical Parameters in Mice

Abstract/ Executive Summary:

The experiment was conducted in 4 groups of mice keeping one control and three treatment groups. Three groups or mice were given dexamethasone (3.5 mg/kg b.wt.). betamethasone (0.5 mg/kg b.wt.) and prednisolone (8 mg/kg b.wt.) to investigate their effects on some haemato-hiochemical parameters. All the mice of the three treatment groups showed a significant decrease in body weight. The blood glucose level was increased significantly in all three treatment groups. TEC, TLC and Hb content were increased significantly in all treatment groups. In case of DLC, monocyte and neutrophil count were increased significantly whereas lymphocyte and eosinophil count were decreased significantly in all treatment groups. It may be concluded that oral administration or glucocorticoids adversely affects some hacrnato-biochemical parameters in mice.

Objectives:

To study the effect of different preparations of glucocorticoids on body weight and haemato-biochemical parameters i.e. TEC, TLC, Hb, DLC, blood glucose in mice.

Methodology:

The research work was conducted in the Department of Pharmacology, Faculty of Veterinary Science, Bangladesh Agricultural University, Mymensingh, for a period of 70 days to study the effects of oral administration of glucocorticoids in mice. The research work was carried out during the period of 1^stAugust, 2001 to 9^th October, 2001. Eight weeks old 20 Swiss albino mice weighing between 25-35 gram were selected for this study. All the mice were maintained under same environment such as similar animal house procedures like feeding, watering, bedding material replacement etc. Three glucocorticoids i.e. Dexamethasone micronised BP, (Oradexon®, Organon Bangladesh Ltd.), Betamethasone BP, (Betnelan®, Glaxo Wellcome Bangladesh Ltd.). Prednisolone USP, (Deltasone®, Renata Ltd.) were used in the present study. The mice were divided into 4 equal groups containing 5 mice in each group. The groups were designated as group A, B. C and D of which group A kept as control. Four types of rations (feed) designated as ration I, 2, 3 and 4 were used in the experiment in which ration 1 was control diet or basal ration. Ration 2, 3 and 4 were basal ration plus dexamethasone, basal diet plus betamethasone and basal diet plus prednisolone respectively. Mice of group A were fed control diet or basal ration. Mice of group B were fed basal ration along with dexamethasone (3.5 mg/kg b.wt) (ration 2). Mice of group C were supplied basal ration along with betamethasone (0.5 mg/kg b.wt.) (ration 3). Mice of group D were fed basal ration along with prednisolone (8mg/kg b.wt.) (ration 4). Body weight and blood glucose level were recorded on day 0 (pre treatment) and on 7^th, 21^st, 42^nd and 70^th day of experimental period (70^th days) by the help of Electronic balance and Glucose meter (Glucotrend) respectively. Haematological parameters were determined on day 0 (pre treatment) and on the 70^th day. The data were analyzed statistically between control and treated values by using well-known 't' test (Paired-Samples T Test) with the help of computer package SPSS.
