Passive Hemagglutination and Complement Fixation Test for the Detection of Duck Plague Virus.

Abstract/ Executive Summary:

A passive hemagglutination (PHA) and complement fixation test (CFT) were carried out to detect Duck plague virus (DPV) or its antibody. In PHA test tannic acid, formaldehyde and glutaraldehyde technics were used, but only tannic acid technic was found sensitive for the detection of DPV or its antibody. It was also found that CFT was sensitive for the detection of DPV or its antibody. This demonstrates that these tests can possibly be used for in vitro detection of DPV and itsantibody.

Objectives:

To find out technic (s) sufficiently sensitive to detect DPV or its antibody. Considering these facts PHA and CFT were tried to determine whether these tests are suitable or not for rapid diagnosis of Duck plague.

Methodology:

Local DPV isolates (DPM-2, DPM-24, DP-180 ) and Duck plague vaccine prepared in France (DPVF) were obtained from the Department of Microbiology and hygiene BAU, Mymensingh. The viruses were propagated separately in chorio-allan-toic membrane (CAM, route in 12 days old duck embryo (DE) for their propagation and these virus isolates were used as antigen in this study. Antisera were prepared using DPM-2, DPM-24, DP-180 and DPVF virus isolates in twelve 4 months old healthy Khaki campbell ducks. Required suspension of Sheep RBC were prepared, following the procedures described and were used both in PHA and CF tests proper. Hemolysin was obtained from central disease investigation laboratory, dhaka and was used in CF test proper, 10% formalin and 1% glutarafdehyde solution these were made according to manufacturer direction and were used in PHA test proper. Glutaraldehyde stabilized pretanned Sheep RBC was prepared by treating with glutaraldehyde following the procedure described with the slight modification. Formaldehyde stabilized pretanned Sheep RBC was prepared by treating with formalin following the method described with slight modification. Tanning of normal sheep RBC, glutaraliehyde and formalin stabilized sheep RBC were done following the procedure. Each type (simply tanned RBC. glutaraldehyde and formalin treated tanned RBC) of tanned cells were sensitized with each type (DP-180, DPM-2, DPM-24 and DPVF) of antigens separately with the procedure. PHA test was conducted with the procedure and with slight modificaticn. Triplicate tests were conducted to make final observation for each of the test technics (Tannic acid technic when Sheep RBC was tanned. Sensitized and used with the antiserum, formaldehyde technic when formalin stabilized Sheep RBC tanned. Achievement/Findings:

This clearly indicated that the viruses and the antiserum used were very specific as a result of which the complement was fixed in the test system resulting non hemolysis in the hemolytic system. The presence of hemolysis in the mixture of DPV and serum dilution of 1:128 and higher was due to the presence of fres complement, which could not be fixed in the test system as a result of insufficient serum, but produced hemolysis in the hemolytic system. The results of the study corelate with that of Lam (1984) who demonstrated lysis of DPV infected DE fibrobfast cell. The results of this study suggest that CFT can be used for in vitro detection of DPV.
