Clonal Propagation of Pineapple Through Tissue Culture

Abstract/ Executive Summary:

Multiple shoots were obtained from dormant axillary buds excised from the crown of pineapple and grown on MS medium supplemented with different combinations and concentrations of NAA, IBA and Kn. The highest number of shoots per explant was obtained from culture grown on medium containing 9.5 uM/l NAA, 9.5 uM/l IBA and 9,5 uM/l Kn. The highest number of roots per plant was obtained when the shoots were grown on medium with 1.0 uM/l NAA and 2.0 uM/l IBA. The regenerated plantlets were successfully established in plastic pots containing soil, sand and cowdung in 1:1;1 ratio.

Objectives:

To develop a protocol for rapid, uniform and large scale production of disease free planting material of Giant Kew pineapple

Methodology:

Dormant axillary buds from crown of pineapple were taken as a source material. Buds were excised with subjacent tissue and washed thoroughly under running tap water and washed again for 10 min with detergent trix. After that the materials were dipped in 0.1 % HgCl₂ solution for 15 min and washed 4 to 5 times with sterilized distilled water to remove all traces of HgCl₂. Explants were prepared aseptically and cultured on MS medium. The pH of all media was adjusted to 5.8. Media were autoclaved for 25 min at 121⁰C and 1.2 kg/cm² pressure. The cultures were kept in growth room at 25⁰C under 16 h photoperiod with a light intensity of 2000 lux under white fluorescent tubes. The materials were subcultured up to five times at 4 to 5 weeks intervals. Then the elongated shoots were separated and individual shoots were placed in the rooting medium. After rooting, the plantlets were removed from the culture flask and washed thoroughly to remove the nutrient agar medium from the roots and transplanted into plastic trays containing a mixture of srerilized soil, sand and well-decomposed cowdung (1:1:1).

Achievement/Findings:

MS medium containing 9.5 uM/l NAA, 9.5 uM/l IBA and 9,5 uM/l Kn was found suitable for in vitro shoot proliferation and half MS medium supplemented with 1.0 uM/l NAA and 2.0 uM/l IBA for rooting of regenerated shoots of pineapple cv. Giant Kew. Thus, the protocol may be recommended for producing pineapple suckers commercially to fulfil the demand of the country but the field performance is still unknown. Though it is vegetatively propagated, it is expected to perform same as the normal suckers.
