Studies on the Mitotic and Meiotic Chromosomes of Sagittaria Sagittifolia L. (butomaceae)

Abstract/ Executive Summary:

Sagittaria sagittifolia L. has 2n= 22 chromosomes in the root tip cells. Their lengths ranged from 4.59 to 15.28 µm with a centromeric formula of 2M+ 2SM + 18AC. Eleven bivalents were formed in most of the pollen mother cells with a mean chiasma frequency of 1.78 ± 0.014 per bivalent. Meiotic irregularities in the form of laggards, micronuclei and failure of cytokinesis were observed in some cells. The analysis of the karyotype and the meiotic behavior indicates that this species in Bangladesh is a basic diploid.

Objectives:

Methodology:

Sagittarta sagittifolia L. is a water loving common weed of rice fields, distributed all over Bangladesh. The materials were collected from the rice fields of Kallayanpur, Dhaka city and also from a dead canal of Mymensingh. These were grown in pots containing clay. Herbaceous plant, propagating by stolon, 25 to 45 cm or sometimes up to 60 cm high, leaves sagittate, 8 to 15 cm long, lobes more or less diverging, petioles 20 to 40 cm trogonous, scape 15 to 40 cm with 3 to 5 whorls of flowers, each 1.5 cm dia, petals white, claw often purple. Feulgen schedule used after pre-treatment for 1 hr in α- bromonepthelene gave the best result for root tip cells (RTCs) although, the chromosome remained little lightly stained. The desired staining intensity could be obtained by adding a drop or so of acetocarmine at the end of the Feulgen schedule. The best preparations of the pollen mother cells (PMCs) were obtained when fresh anthers were squashed in 2% acetocarmine. The peak period for metaphase I (MI) was between 9 and 11-30 A.M.

Achievement/Findings:

Since most of the somatic chromosome pairs are homomorphic and a high average chiasma frequency per bivalent are present, this species in Bangladesh can be regarded as a basic diploid. The meiotic abnormalities observed in this study are possibly due to one or more kinds of structural aberration in its chromosomes, for understanding the exact nature of it further study is needed.
