Effect of Varieties and Growth Regulators on in Vitro Regeneration Potentiality of White Jute (corchorus Capsularis L.)

Abstract/ Executive Summary:

An experiment was conducted to investigate the effect of varieties and growth regulators on in vitro regeneration of white jute. Cotyledon of six varieties of white jute namely CVL-1, CVE-3, CC-45, BJC-7370, D-145 and C-83 were cultured on different concentrations of growth regulators for callus and subsequent regeneration. Seed germination was better in variety CVE-3 (77.08%) with cotton-based MS medium rather than agar based medium. The highest percentage (97.14%) of callus induction was found in the combination of CVE-3 with 0.5mgl-1 IAA and 2.0mgl-1 BAP followed by CVL-1 (88.57%) in same growth regulators concentration. The highest shoot regeneration (73.33% was found in variety CVE-3 with 0.5mgl-1 NAA and 2.0 IBA. After root initiation the regenerated plantlets was transferred to soil. The establishment of rooted plantlets in the field soil was satisfactory. The regeneration ability of these varieties has the potential for use in future biotechnological studies and genetic improvement of Corchorus capsularis.

Objectives:

The study was undertaken to develop a more efficient in vitro plant regeneration system for C. capsularis which may be used for successful regeneration through genetic transformation.

Methodology:

The experiment was conducted during the period from February to October, 2007 in the Biotechnology laboratory of Bangladesh Agricultural University. Mymensingh. Six cultivated varieties, of white jute namely CVL-1, CVE-3, CC-45, BJC-7370, D-145 and C-83 were used for the study. Seeds were germinated in vitro followed the method (Huda et at., 2007). In vitro grown Cotyledon of six varieties was used as explants. The explants of six varieties were then cultured on MS (Murashige and Skoog, 1962) medium supplemented with various concentrations and combinations of IAA (0.1, 0.5 and 1.0 mgl-1) and BAP (1.0, 2.0 and 3.0mgl-1) for callus induction. For shoot regeneration, calli were cultured on MS medium supplemented with various concentrations and combinations of NAA (0.1, 0.25, 0.5, 0.75 and 1.0mgl-1) were used for root induction. The pH of the medium was adjusted to 5.8+0.1 before addition of 0.8% (w/v) agar for solidification. All the cultures were maintained in 16 h photoperiod, with light intensity of 2000-3000 lux at 25+10C. Subcultures were carried out as and when required. Plantlets with sufficient roots were transferred to small plastic pots containing sterilized soil. These plantiets were acclimatized and then transferred to the field. The experiment was conducted in Completely Randomized Design with five replications. Analysis of variance for different component was computed and the difference between means was evaluated by Duncan’s Multiple Range Test (Gomez and Gomez, 1984.)
