Plant Regeneration from the Cotyledon with Attached Petiole of Six Varieties of White Jute

Abstract/ Executive Summary:

Six varieties of white jute (CVL-1, CVE-3, CC-45, BJC-7370, D-154 and C-83) and different combination and concentrations of grown regulators were used to investigate their in vitro regeneration potentiality. Variety CVE-3 with cotton based medium was found to be the higher seed germination (82.0%) than the variety BJC-7370 with agar-based MS medium with (53.0%). Maximum callus (100%) induction was found in the variety CVE-3 with 0.5mgl-1BAP. The highest shoot regeneration (80.0%) was observed in the same combination. The best root initiation (86.66%) was found in the variety CVE-3 with 0.25mgl-1 IBA. The in vitro grown plantlets were successfully transplanted into soil.

Objectives:

To develop a reproducible and efficient protocol for in vitro regeneration of six varieties of white jute.

Methodology:

The experiment was conducted in the Biotechnology Laboratory, Bangladesh Agricultural University from February to October, 2007. Seeds of the xix cultivated varieties, namely CVL-1, CVE-3, CC-45, BJC-7370, D-145 and C-83 of white jute (Corchorus capsularis L.) were surface sterilized by immersing in absolute alcohol for I minute and then in 0.1% (v/v) Mercuric Chloride for 20 min, followed by 6 washes with distilled water. Seeds placed on the surface of agar-solidified (0.8%, w/v) and cotton based MS medium for seedling production. Cotyledon with attached petiole of six varieties C. capsularis were cultured on MS medium with various concentrations and combinations of IAA (0.1, 0.5 and 1.0 mgl^-1), BAP (1.0, 2.0 and 3.0 mgl^-1) and NAA (0.1, 0.25, 0.5, 0.75 and 1.0 mgl^-1) for callus induction and subsequent regeneration. Shoots of xix varieties C. capsularis again cultured on MS medium with various concentrations of IBA (0.1, 0.25, 0.5 0.75 and 1.0mgl^-1) for root induction. After sufficient development of shoot and root systems, the small plantlets were acclimatized and transplanted to small pots containing sterilized ground soil mixed with cowdung. The experiments were arranged in Completely Randomized Design with 5 replications. All data were statistically analyzed and mean difference was evaluated by Duncan’s Multiple Range Test (Gomez and Gomez, 1984).
