Tissue Specificity and Comparative Esterase Variability in Notopterus Notopterus and N. Chitala (osteoglossiformes: Notopteridae)

Abstract/ Executive Summary:

Investigation was carried out on the expression pattern of non specific esterases (on 7.5% PAGE with α naphthyl acetate) from ten different tissues of Notopterus notopterus and N. chitala, indigenous to Bangladesh. Altogether four esterase bands (Est-1, Est-2, Est-3 and Est-4) were found in both species showing tissue and species-specific expression. Est-1 was common in all the tissues of both species with different staining intensities. Est-2 was found in the liver and heart of N. notopterus and in the brain, liver and stomach of N. chitala. Est-3 was detected only in the stomach of N. notopterus but in the liver and midgut of N. chitala. Est-4 was detected in the eye of N. notopterus and in the liver of N. chitala in addition with the brain of both species. The frequencies of esterase bands were comparatively higher in the tissues of digestive and nervous system of both species. In general, N. chitala had higher esterase concentration than in the N. notopterus.

Objectives:

To find out the tissue specific esterase isozyme banding pattern in two species of Notopterus and comparing of the banding pattern as molecular marker to identify them, based on suitable tissues.

Methodology:

Samples were collected from Ariyal Beel (Mawa Fish Market, Munshigonj) and the experiments were conducted in the laboratory of Genetics and Molecular Biology, Department of Zoology, University of Dhaka. Ten (pelvic_, anterior_ and tail_muscle, foregut, midgut, eye, brain, stomach, heart and liver) and five (later five ones) different tissues (nearly 0.016 g) were taken to observe the tissue specific esterase isozyme variability and to compare the banding pattern from both species respectively. Each tissue was squashed in TBE buffer (40 μl) and aliquots from each sample (15 μl) were loaded on the gel slots for electrophoresis after centrifuged at 12500 rpm for 15 min. The entire technique for polyacrylamide gel electrophoresis (PAGE) was followed and the electrophoretic bands of esterase isozymes resulting from stained gel with α naphthyl acetate were assigned toincreasing numbers based on decreasing mobility. The experiment was repeated to standardize the result with different specimens. As there were no significant variations in each repetition, only one repetition was subjected to analysis.

Comparative study revealed that esterase isozymes banding pattern in different tissues of the two species showed both similarity and dissimilarity in their staining intensity as well as in the occurrence of the number of bands. Differences in the number of bands between species may have underlying mechanisms regulating the esterase-related processes. Expression of esterase isozymes could be used successfully as a marker to understand localization of this enzyme in different tissues of a species that could also be used any conservation effort as well as species identification and selection.
