Comparison of Tissue Specific Esterase Isozyme Banding Pattern in the Larvae and Adult of Heteropneustes Fossilis

Abstract/ Executive Summary:

The variation of esterase isozymes expression from nineteen different tissues of stinging catfish (Heteropneustes fossilis) was studied by 7.5% polyacrylamide gel electrophoresis (PAGE) after staining with alfa- and Bita-naphthyl acetate substrates. Two stages, larva (without gonad) and adult (with gonad) were taken for the study. Maximum four esterase bands namely Est^-11.45, Est-^21.22, Est^-31.00 and Est^-40.23 were observed in both the larvae and adult. The bands showed tissue specific distribution and varied in their staining intensity. Est-1 was observed only in liver and kidney of the larvae and caudal fin of the adult. Est-2 was more frequently observed in adult tissues (liver, kidney, bile, stomach, intestine, rectum, heart, gill, fore- and hind-brain, caudal-, ventral- and pelvic-fin) than those of the larvae (liver, intestine, dorsal muscle, heart, gill, fore-, mid- and hind-brain). Est-3 was common in all of the tissues in both the larvae and adult except in the gill of adult. Est-4 was found in some tissues of the larvae (liver, kidney, dorsal muscle, brain and caudal fin); however, it was more frequently observed in the tissues of the adult (liver, kidney, stomach, intestine, rectum, heart, brain and caudal fin). Relatively higher concentration of esterase isozymes were found in the digestive and nerve tissues.

Objectives:

To investigate the localization of esterase isozymes expression in ?different tissues of adult stinging catfish and its various larval stages to understand the changes of esterase isozyme electrophoretic patterns with aging.

Methodology:

Fish samples were collected from Buriganga river (latitude 90⁰25’N, longitude 23⁰45’E) and the experiments were conducted in a laboratory of the University of Dhaka. Esterases were identified in the gels following the technique by using a mixture of both alfa- and bita-naphthyl acetate substrates. The entire technique for polyacrylamide gel electrophoresis (PAGE). Adult samples weighed about 60 g and attained 29 cm in length, while the larvae weighed about 5 g and attained 10 cm in length. Nineteen different tissues namely liver, kidney, bile, stomach, intestine, rectum, anterior dorsal muscle, mid dorsal muscle, posterior dorsal muscle, eye, heart, gill, fore brain, mid brain, hind brain, tail fin, dorsal fin, ventral fin and pelvic fin were taken from both the larval and adult stages. For each test, nearly 0.0159 g tissue was taken. These were separately squashed in TBE buffer, centrifuged at 12500 rpm for 15 min and aliquots from each sample were loaded on the gel slots for electrophoresis. The experiment was repeated several times with different specimens of both stages. There was no significant variation of esterase isozyme banding pattern for each repetition, so only one representative gel was subjected to analysis to identify bands on gel.

Achievement/Findings:

The expression of tissue specific esterase isozymes in the larvae and adult of the stinging catfish showed variations, i.e., different banding patterns. The results may be useful in future studies. Esterases could also be used as bioindicators to measure the toxic potency of pesticide residues in catfish habitats.
