Tissue Specific Esterase Isozymes Banding Pattern in Silver Carp (hypophthalmicthyes Molitrix) at Different Developmental Stages

Abstract/ Executive Summary:

The electrophoretic banding pattern of esterase insozymes were examined on 7.5% polyacrylamide gel electrophoresis (PAGE) stained with alfa and bita-napthyl acetates in 17 different tissues of Hypophthalmicthyes molitrix from three different stages (5 & 8 months old larva-without gonad and adult-with gonad). Altogether six esterase bands (Est-1^1.93^±^0.04, Est-2^1.63^±^0.04, Est-3^0.99^±^0.03, Est-4^0.68^±^0.03, Est-5^0.34^±^0.04 and Est-6^0.16^±^0.04) were observed where Est-3 and Est-4 were found to be more frequent. Est-6 was rarely present in all the tissues and was totally absent in the 5 months old larva. All six bands were appeared only in liver tissue. Banding pattern was more or less same in the different parts of eye, brain and muscles in all three stages but varied from tissue to tissue. Comparatively higher esterase activity both interms of staining intensity and occurance of the number of bands was found in digestive and nerve tissues which might be used for the further study of physiology, taxonomy and toxicology.

Objectives:

To understand the changes of esterase isozyme patterns with aging to ensure the better growth rate, high yeildings and also for taxonomical analysis.

Methodology:

Samples were collected from open water body near to Dhaka city (N-90⁰25’, E-23⁰45’) with the help of a fisherman and the experiments were conducted in the laboratory of Genetics and Molecular Biology, University of Dhaka. Seventeen different tissues (nearly 0.0159 g) namely anterior muscle, mid dorsal muscle, tail muscle, eye (black), eye (lens), fore gut, mid gut, hind gut, liver, kidney, fore brain, mid brain, hind brain, heart, intestinal bulb, bile and gall bladder were taken and squashed in TBE buffer (40 μl) from both larval and adult stages. Aliquots from each sample (15 μl) were loaded on the gel slots for electrophoresis after centrifuged at 12500 rpm for 12 min. The entire technique for polyacrylamide gel electrophoresis (PAGE) and the electrophoretic bands of esterase isozymes resulting from stained gel with α_and β_naphthyl acetates were assigned to increasing numbers based on decreasing mobility. The experiment was repeated for two more times with different specimens and little variation of esterase isozyme banding pattern was found in each repetition, so only one repetation was subjected to analysis.
