Bacteriological Quality of Tanualosa Ilisha (hamilton) in a Freezing Plant

Abstract/ Executive Summary:

Bacteriological analyses were carried out to determine the quality of Tanualosa ilishe in a freezing plant.The viable cell counts of different groups of bacteria in the flesh of frozen hilsha fish, unfrozen hilsha fish and related environments were determined by standard methods. The total viable bacterial count, Coliform and faecal coliform in the flesh of unfrozen Hilsha fish were found higher than that found in the fresh of frozen hilsha fish. From the experimental findings it was shown that the standard plate count(SPC), Coliform and faecal coliform in case of unfrozen hilsha fish did not comply with the International Commission on the Microbiological Specification of Food (ICMSF). The bacterial loads in freezing frozen hilsha fish complied with ICMSF standard. But one lot of frozen hilsha fish contained 15 faecal coliforms per gm. This finding suggests suggest that it is very necessary to check frozen fish for faecal coliform at regular intervals. Vibrio cholerae and Salmonella spp. were not detected in all samples studied.

Objectives:

To know the total bacterial load of frozen hilsha of a Snowing freezing plant and unfrozen hilsha of different fish markets.

To know the total coliform and faecal coliform of the same species of the same place. To know the detection of salmonella and also the detection Vibrio cholerae for the assurance of quality of frozen and unfrozen hilsha for export.

Methodology:

Twelve frozen hilsha samples were collected lot wise from Snowing freezing plant , Mirpur, Dhaka and twelve unfrozen hilsha fishes were collected from Chandpur and local markets of Dhaka.Water samples were collected from taps and panning table of freezing plant and flake ice samples were collected from ice store of the freezing plant during the collection of fish samples and carried to the Laboratory of Fish Inspection and Quality Control Laboratory, DoF, Dhaka.for analyses. Moreover swab samples were collected from the surface of the panning tray and the surface of workers hands of the freezing plant.Total plate count was determined by ten fold serial dilution method and calculated as cfu/ml. Coliform and faecal coliform counts were determined by Most Probable number Method. Detection of Salmonella and Vibrio cholera were done by using respective agars.

Achievement/Findings:

The results of the total microbiological loads, total coliform and faecal coliform of the frozen hilsha in freezing plant comply with ICMSF standard.On the other hand the same results of the unfrozen hilsha of the two markets like New market and Kawran bazar don't comply with ICMSF standard.But the same results of the other two fish markets comply with ICMSF standard.Vibrio cholerae and Salmonella spp. were absent totally in all the samples. SPC of the frozen fish was lower than that of unfrozen Hilsha fish collected from different markets and fish habitats.The SPC of water found to be varied in the range of 226.5/ml to 1630 /ml. In case of flake ice the SPC varied between 595 /ml and 1360/ml Coliform was found in flake ice 25/ml The SPC of Swab was 195/cm square, and the same of the hand of the panning worker was 225/ square cm. It was observed that in case of all samples Spc was higher than the recommended by EU standard. So, it should be checked properly in the processing plant.
