Comparative Study of Four Locally Isolated Strains of Rhizobium Japonicum.

Abstract/ Executive Summary:

The experiment was conducted to make a comparative study of four promising locally isolated strains of R. japonicum against Nitrogen. The soybean variety used f or the study was Bragg. The experiment was laid out in a randomized block design with three replications. The performance of the strain, GII-5 was found to be as good as that of Nitrogen and better than that of the other strains under study.

Objectives:

To study of the effectiveness in field conditions on the growth of plant and yield of soybean.

Methodology:

The investigation was made in the field laboratory of BCSIR, Dhaka during January to April, 1980. Four promising locally isolated strains of Rhizobium japonicum GI-13. GII-5, GII-7 and 0-4, one commercially used inoculum "Nitrogen' and a control were included in this investigation. The purified strains were cultured and preserved in slants of yeast mannitol agar having the following composition (per litre) : Dipotassium hydrogen phosphate. 0.5 gm; magnesium sulphate 0.2 gm; sodium chloride 0.1 gm; mannitol 10.0 gm yeast extract 2.5 gm and agar 20 gm. For propagation mannitol was replaced by 0.5 % sucrose in the above mentioned medium. The shake culture method was used at room temperature and shaking was continued for seven days. The cells were harvested by centrifugation and put into sterile peat carrier (peat 40%; garden soil 54.5%. ; calcium carbonate 5% and sucrose 0.5%). The carrier containing the strains was air dried at room temperature and preserved in polythene bags. The soybean variety, Bragg was used for the study. The experiment was laid out in a randomized block design with 3 replications. The unit plot size was 3m x I .5m. Two to three seeds were put in each hill of 7.5 cm. distance within the rows which were spaced 30cm. apart and after germination the hills were thinned to single plant. The inoculations were done by treating the seeds with 1% gum acacia solution and then mixing thoroughly and uniformly with inoculum (12 gm/kg of soybean seed). This treatment was done just prior to sowing and after sowing the seeds were covered with soil to protect them from sunlight. Ten plants were selected at random from each plot to have data on nodule number and dry weight of nodules at 30 and 60 days and fresh weight of nodules and bacteroid number at 60 days. To determine the bacteroid number the collected nodules were weighed and surface sterilized by following the method of Vincent. The viable counts were determined according to the plating method of Vincent using yeast mannitol agar with congored as plating medium. At har vest, data on plant height were obtained form ten randomly selected plants from each unit plot.

Achievement/Findings:

Maximum plant height was recorded from Nitrogen and GII-5 followed by GIl-7 but without any statistical difference. The shortest plants were recorded from the control. The highest seed yield was recorded from GII-5 closely followed by Nitrogen but they did not differ statistically from one another, GI-13 and 0.4 gave identical yields. The lowest yield was obtained from GIl-7. It may be mentioned that the highest yield due to the strain, GlI-5 was associated with more number and weight of nodules per plant and higher plant height. So this strain can be used for economic benefit of soybean cultivation.
