To Investigation the Sensitivity of Sclerotium Oryzae to Various Chemicals

Abstract/ Executive Summary:

The in-vitro effect of three fungicides (Dithane M-45, Dithane Z-78 and Formalin) and three herbicides (Baspapon, Tokgranule and Ramrod) against Selerotium oryzae was evaluated by soil-drench and soil-mix methods. In both tests, hundred per cent inhibition of sclerotial germination was obtained with formalin at a concentration as low as 1000 ppm. At higher concentrations (5000 and 6000 ppm) however, some amount of inhibition was obtained by Dithane M-45 in soil-mix method. None of the herbicides could affect the viability of sclerotia in any of the tests even at 6000 ppm.

Objectives:

To determine the Sensitivity of sclerotium oryzae to Various Chemicals.

Methodology:

The merits of the sensitivity of the chemicals were evaluated in the laboratory on their relative effect on the viability of sclerotium. In general Zentmyer's soil-drench and soil-mix methods with modifications were followed. Fine, air-dried and seived, sandy loam soil was autoclaved for 45 minutes at 15 Ibs pressure. After a week, test tubes were filled with this soil upto a depth of one inch. Small discos of mycelium with sclerotia were cut from 10-day old culture. A single disc was placed on the soil in the tube and covered by one inch of sterilized soil. A sample of chemical solution/suspension sufficient for soaking the two inches soil (5 ml) was poured on the top of the soil. The tubes were incubated at 30°C. After 24 hours, the disc was collected and the soil particles were removed by gentle washing. Sclerotia were then aseptically transferred to PDA plates for germination. The chemicals were tested at six concentration (1000,2000, 3000, 4000, 5000 and 6000 ppm) using three replications with distilled water control. Steam sterilized soil was mixed with a measured amount of chemical and test tubes were filled upto a depth of one inch. A mycelia disc with sclerotia was placed on the soil and covered by another inch of sterilized soil. A sample of 5 rnl sterilized water was added to the soil and the tubes were plugged. After incubating 24 hours at 30°C the disc was removed and the sclerotia were transferred to PDA plates aseptically. Each treatment was replicated thrice with no chemicals in the soil as control. The per cent inhibition of sclerotiaI germination was determined as in the preceding test.
